The Isolation and Identification of Porcine Parvovirus in Sichuan region Xu Jing Abstract One Porcine Parvovirus strain is isolated from the stillborn of sow in Sichuan through pig kidney cell and established cell lines (Swine testis, SI). It is identified through morphology, serology, physical and chemical characteristics, cytopathic effects (CPE), inclusion bodies, Polymerase Chain Reaction(PCR) and so on. It shows that the porcine parvovirus is susceptible to 5-bromouracil deoxyribose and can resist ether, chlorofonn, pH 3.0, 56C 30mm and trypsin. It can agglutinate the RBC of guinea pig9 chicken, which is inhibited by the positive serum. It produces intranuclear inclusions within 20-24hours after inoculation. The virus produces pronounced cytopathic effects, and the effects can be neutralizated by the positive serum. Under electron microscope there are 22 ?2 nanometer virions, which are similar to the standard ones in structure. Swine testis cell lines is more suitable than IBRS-2 to propagate the virus. The serotype of the virus is the same as the other strain PPV through cross-agglutination. Doing PCR with Extracted the virus replication form DNA, the result shows there is a 2.0kb band as the other strain PPV. All of these demonstrate that the virus has the same character as the PPV. So it is proved that the virus is a strain PPV.
