In this experiment,ears,blood and the semen in part of samples were randomly organizations, build the sheep breeds gene pool; using AFLP molecular markers for three local sheep breeds to genetic diversity analysis, AFLP technology is the first time used by the Xinjiang local sheep breeds applications. The results as follows:1.Investigate the situation for Cele sheep,Duolang sheep and Kerkez sheep for the genetic breed.Cele sheep were breed establish conservation areas in Cele city, the implementation of self-breeding group,enhance breeding in the region,consolidating and enhancing the performance of multiple births.Duolang Sheep breed mainly in Kashi Prefecture Maigaiti city.conservation through the live animals and frozen semen preservation the method of combining,The conservation work carried out at the same time to strengthen the breeding.There is no corresponding areas of Kerkezi sheep,there is no interference by the exotic species,therefore,it can be established as soon as possible for protection of this species.2.The corresponding treatment of the ears,blood.then establishment of the three sheep breeds organization, gene pool of blood.Taken Cele sheep,Duolang sheep and Kerkezi sheep's blood and ear organizations taken into the eppendorf of 1.5ml centrifuge tubes,stored in liquid nitrogen,tissue gene pool;the blood samples taken by the sub installed after the extraction of DNA carried out in part to the diversity of analysis,the part of remaining take into the 10ml glass packaging,to preservation of the ultra-low temperature refrigerator, build blood genome.3.Genomic DNA was prepared by Sambuck,and the size of 50kb genomic fragment, in order to lay a foundation for AFLP optimized system in next experimentit can be identified with a clear, strong signal amplification, band-type stability, AFLP suitable for analysis of genetic diversity of sheep.4.Screened with the 11 pairs of E+3/M+3 primer, the 3 local breeds of sheep genomic DNA for AFLP amplification. According to the results of AFLP analysis, statistics for each primer combination was detected in all species of polymorphic bands,and the specific bands were calculated in three sheep,similarity and genetic distance and UPGMA clustering to build,the relationship of Fig.11 pairs of primer combinations a total of 543 clearly identifiable bands,per primer combinations generated 49.37 polymorphism, polymorphic bands for the 54, the average percentage of polymorphic loci was 9.94%, while the varieties also detected specific bands.5.The UPGMA cluster analysis of the adoption will be three local sheep breeds are divided into 2 populations.Three local sheep breeds have been a certain degree of genetic diversity,their genetic similarity coefficients and clustering results with the geographical distribution of sheep breeds, coincide with the actual situation.
