| Pigeon pea [Cajanus cajan(L.)Millsp.)] is one of the important tropical crops and has high medicinal value.Predecessors have little research on genetic diversity and medicinal resources of pigeonpea germplasm resources.In this study,69 pigeon pea accessions(cultivars)at home and abroad were used as materials,and their genetic diversity was analyzed using morphological markers and SSR markers.The effect of salt tolerance of pigeon pea is preliminarily explored,and its medicinal value is evaluated by the determination of total flavonoid content and main medicinal components,in order to provide a basis for the selection and application of new varieties of pigeon pea.The main findings as fellow:1.Morphological diversity analysisThrough the determination and analysis of 10 quality traits and 14 quantitative traits of 69 pigeon pea germplasm resources,the results show that there are significant differences in morphological traits between different pigeon pea germplasm resources,and the genetic diversity index of 10 quality traits is relatively high.Ranged from0.546 to 1.000;the coefficient of variation of 14 quantitative traits ranged from 7.75%to 23.39%.Correlation analysis showed that,except pollination type and other quality traits are not related,the other 9 quality traits are significantly related to other traits,and insect resistance is significantly related to growth habit,leaf shape,stem color and seed stain.The number of seeds per pod was significantly positively correlated with flowering period,number of single-branch inflorescences,pod length,pod width and grain length,and significantly negatively correlated with leaf width and grain width.Based on the cluster analysis of 6 important traits,69 pigeon pea germplasm resources can be divided into three categories,that is medium-grain late maturity,large-grain medium-early maturity,and small-grain medium-early maturity.2.Analysis of genetic diversity of SSR molecular markersThe 42 polymorphic SSR markers selected were used to analyze the genetic diversity of 69 pigeonpea germplasms.A total of 118 alleles were detected,with 2 to5 alleles per marker,with an average of each marker 2.8.The genetic similarity coefficient values ??among 69 pigeonpea germplasm resources are mostly distributed between 0.550 and 0.650,accounting for 59.36% of the total number of detections;through cluster analysis,69 pigeonpea resources are divided into 4 major resources at the similarity coefficient of 0.578.3.Preliminary evaluation of salt tolerance of excellent germplasmSelect 5 pigeon pea from different sources and observe the seed germination characteristics and seedling growth of different concentrations of Na Cl solution.It is found that Na Cl stress can significantly inhibit seed germination and seedling growth.During the seed germination and seedling stage,5 pigeon pea tolerant of salt ability is not completely consistent.MD1 and GX1 are salt-tolerant germplasm,and YN6 is salt-sensitive germplasm.During the germination and seedling stages,the critical concentrations of Na Cl stress for 5 pigeonpea were 0.377-0.748% and 0.275-0.510%,respectively.4.Evaluation of effective ingredientsContent of total flavonoids,stilbene acid and 6 flavonoids of 69 pigeonpea germplasm resources were analyzed by ultrasonic method and ultra-high-performance liquid chromatography-triple quadrupole mass spectrometry(UHPLC-QQQ-MS/MS),respectively.Measurement results showed that the total flavonoid content of 69 different pigeonpea germplasms was significantly different,and the variation range was 1.18%~10.98%.There are significant differences in the content of the effective components between different pigeonpea germplasms,and there is a significant correlation between the effective components.Naringenin and quercetin,vitexin,genistein and luteolin are all significantly positively correlated(P<0.01).According to the cluster analysis of the medicinal components,69 pigeonpea germplasm resources can be divided into 4 categories.Medicinal value of 69 pigeonpea germplasm resources was comprehensively evaluated by the membership function method,and the highest and the lowest 5 pigeonpea resources were screened out. |
