| Red tide is one of marine disasters all over the word. The flow imaging cytometry is a device using both flow cell technique and flow imaging technique. Through analysis and recording the types and amount of the planktons in seawater, the development and tendency of red tide can be monitored under the help of this machine. A prototype of optical system based on the objective of biological microscope had been established in this thesis and a series of experiments have been done to improve the system.Firstly, The emission spectrum and excitation spectrum of several kinds of planktons had been measured to provided experimental evidence for excitation light source choosing; a new pattern of fluorescence excitation and detection had been used. Secondly, a series of illumination experiments had been finished to certificate the prototype of optical system; critical illumination effect was to some extent fit to use; the different illumination effect under LED with different wavelengths had been explained theoretically and in practice. Finally, a new microscope lens had been designed. |
