Abstract/Summary:
The paper used tomato sauce which was produced in Xinjiang as raw material to optimize the best conditions of tomato sauce saponification using sodium carbonate as the base. The organic solvent extraction, ultrasonic extraction and microwave extraction were studied as the three extraction methods, the best extraction conditions of each method was developed. Meanwhile, dried tomato powder was used as raw material to study the entrainer and the best extraction condition about using supercritical carbon dioxide to extract lycopene.The purification technology of lycopene oleoresin was studied by using column chromatography. The lycopene crystals were prepared by column chromatography and later recrystallization. Thin layer chromatography, melting point test , Ultraviolet absorption spectrum, Infra-red spectrum and high efficiency liquid chromatography were used to characterize the lycopene crude extract and lycopene crystal. By doing this, some useful manufacturing parameters and technical parameters can be provided.This paper also focused on the stability of lycopene and found some influencing factors affecting its stability in order to provied some useful reference data for lycopene products' color care and safe storage. In vitro anti-oxidation experiment was performed in the last part of this paper. Hydroxyl radical scavenging activity and hydroxyl radical scavenging activity by lycopene were tested, using the same concentration of L-Ascorbic acid as the control group. The results showed that lycopene was a good free radical scavenger for both the free radicals.
