The subject researched the extraction-purification technology of total flavonoids in Speranskia tuberculata,and established a method of determination of flavonoids in Speranskia tuberculata and its effective parts.Preliminaryily established quality standards of effective parts of Speranskia tuberculata.In extraction technology process,two methods of extraction were studied extract the total flavonoids in Speranskia tuberculata,The result of refluxing method to extract the total flavonoid was better,so we choose to use a refluing method.We investigated the influence of four factors:density of ethanol,the quantity of extraction solvent,extraction time,extraction number of times to extraction technology,In the end,we determined the optimum extraction technology of flavonoids in Speranskia tuberculata parameter is:70%ethanol as solvent,the ratio of material and solvent is 1:12,extracting three times with 1h for each time at 75℃.In purification technology process,through sieving,we use macroporous adsorption resin to purify the flavonoids in Speranskia tuberculata.In the end, we determined the optimum purification technology of total flavonoids in Speranskia tuberculata is:take the sample by 1BV/h for 2 times,after adsorbing 12h,eluting with 5BV H2O by 2BV/h,at last use 5BV 60%ethanol by 4BV/h.We determined the content of A in Speranskia tuberculata.Results showed the method had a good linear range,accuracy,stability,responsibility and recovery, and may serve as a way of quality stantard drawing up.Through the DPPH antioxidant activity test,we can found that total flavonoids in Speranskia tuberculata have a certain antioxidant activity,are similar to VE.
