| The prescription of Tong feng is a proved clinical effective recipe which had been used several years by old TCM experts. It consists of Rhizoma Corydalis, Cortex PhellodendriChinensis, Rhizoma Atractylodis Macrocephalae and so on. It is beneficial for treating rheumatoid arthritis due to syndrome of heat-dampness. It has the effect of clearing heat and eliminating dampness, invigorating blood circulation and dredging channels and collaterals, relieving pain. My research was based on clinical soup dosage and processed study, by prescription analysis and dosage form selecting, and prepared tongfeng granulea. The research dealed with preparation process study and quality standard study in order to research a safety, convenience, and effective newmedicine which basic curative effect remained.The main contents of this paper were as following:Preparation process study:Optimization of the influence of the mixed volatile oil extracting was chosen the crude drugs broken the pieces, quality of water and time of extracting. We select the optimal procession by L9(3)4 orthogonal test. Results showed that extracting must elect Atractylodis Macrocephalae with 6 times water, extracting 8 hours and smashed it into 20 mu.Optimization of the influence of the alcohol-extracting of Rhizoma Corydalis was chosen the extracting method. We used and Tetrahy -dropalmatine as index to select the optimal procession by L9(3)4 orthogonal test. The best conditions for alcohol—extracting were adding 80% alcohol 6 times, heating circumfluence 3 times for 2 hours Per timeOptimization of the influence of the refinement of Cortex Phellodendri Chinensis and orther drugs was chosen of density of clinical soup, time of alcohol-immersing and time of placing. We used contents of dry extract and the Berberine Hydrochloride as index to select the optimal procession by L9(3)4 orthogonal test. The best conditions for the refinement were that the density of clinical soup is2:1, time of alcohol-immersing is 60%, time of placing is 12h.Optimization of the processing of mixed volatile oil which wrapped byβ-cyclodextrin(β-CD) was chosen the utilization ratio of mixed volatile oil as the index. The factors affecting wrapping procession were researched by L9(3)4 orthogonal test. The best conditions for mixed volatile oil which wrapped byβ-cyclodextrin(β-CD) were containing oil andβ-CD with a ratio of 8∶1 for 120 minutes at 30℃.Quality standard study:Rhizoma Atractylodis Macrocephalae and Medical A, B, C in tongfeng an granulea were identified by TLC. Tetrahydropalmatine and Berberine Hydrochloride in tongfeng an granulea was determined by HPLC. The results showed that the stability, the rate of recovery, reproducibility and accuracy were satisfactory. Tongfeng an granulea was satisfied with all requirements of granulea in the 2005. edition C.P.(the first hart). |
