Pyrola is the dry herbs of Pyrola callitha H.Andres or Pyrola decorate H.Andres. Pyrola contains many kinds of active components just as hyperin, 2"-O-gollaol-hyperin, homoarbutin, pyrolin, 6'-O- gollaol- homoarbutin and so on. Modern Pharmacology manifests that Pyrola can increase immunity of organism, broaden blood vessel, advance circulation of aeteria coronaria, cut down blood pressure, adjust blood fat and so on. Using Pyrola to cure heart and cerebrovascular disease gains certain curative effect.It has great prospect of development. This paper adopts macroporous adsorptive resins to purify the active ingredient of total flavonoids and total polyphenols in Pyrola. In order to break new way of curing heart and cerebrovascular disease by traditional Chinese medicine, we use the technology of solid dispersion to prepare Pyrola dropping pills, which have quick results and high bioavailability.Taking the extraction yield of total flavonoids and total polyphenols as index, we compare the methods of ethanol ultrasonic, ethanol circumfluence and water circumfluence.In the end, we choose ethanol circumfluence.we determine the extraction technology of Pyrola by orthogonal experiment. The optimum extraction procedures are as follows: extracted with 10 double times 50% ethanol circumfluence for 3 times and 1 hour each time.This paper using macroreticular resin purify Pyrola extractum and optimize purification process. The best technics are as follows: the type of macroreticular resin is LSA40, the sample fluid is densed to (1:10), the flowing rate is 2.5 BV/h , the throughput of LSA40 is 1g Poyrola per mil, the water is 5 BV, the eluant is 2. 5 BV 70% ethanol, the flowing rate is 2BV/h . The extractum contains 22 percent total flavonoids and 9 percent total polyphenols.We establish UV methods for determination of total flavonoids and total polyphenols and a HPLC method for determination of hyperin in Pyrola's extractum and dropping pills.They can be used to control the quality of the Pyrola dropping pills.The results of methodology tests prove that these methods are all accurate, stable and reliable.Based on the results of preliminary test and orthogonal experiment, we determine the prescription of the Pyrola dropping pills.The results are as follows: using PEG4000/PEG6000=1:1 as matrix, extractum/matrix=1:2.Taking appearance score and weighting varitation as index, we study on the influence factors: the temperature of melting liquid, the temperature of condensate, the inside diameter of drop head, the dropping distance by orthogonal experiment. The results are as follows: the melting temperature is 85℃, the condensate is silicon oil, the temperature of condensate is 0℃, the inside diameter of drop head is 2cm, the dropping distance is 10cm. This paper studys on the quality specification of Pyrola dropping pills.We establish the specification in many ways, such as character, TLC identification, weight difference, content determination and so on. The pill's state of solid dispersion is evaluated by the study of X-Ray diffraction analysis and vitro dissolubility.We also evaluate the dropping pill's stability. The results manifest that the drug exists in the dropping pills by the state of microcrystal, and that the time of resolution is short, and the dropping pill's quality is controllable and stable.
