Pipeline in Piper nigrum Linn were identified by TLC with clear and reproducible spot, the content of pipeline in extractor was determined by HPLC. Active components showed good linear relationship in the range of 0.01μg-0.1μg(r=0.9999). And the average recovery was 98.67%, RSD=1.15%(n=9). The two methods can be used for the quality control.By using the weight of pepper oil and the content of pipeline as quality control criteria, The effects of pressure, temperature, extraction time, the flow rate of CO2, and granularity of material on the extractive results of Piper nigrum Linn were explored at the same time, the curves of the influence factors were drawn. The SFE- CO2 conditions for pepper oil were as followed: pressure 10MPa, temperature 35℃, the rate flow of CO2 2.4L/h, extraction time 3h, the granularity of material was 60. The result showed that the oil extractive yield was 1.73%.By using the content of pipeline in extractor and the extractive yeild of peperine as quality control criteria, The effects of pressure, temperature, extraction time, the kind of modifier and the flow rate of modifier on the extractive results of Piper nigrum Linn were explored at the same time, the curves of the influence factors were drawn. The SFE- CO2 conditions for piperine were as followed: pressure 30MPa, temperature 60℃, extraction time 3h, the modifier was 95 % ethanol, the ratio of the the material and the entraner was 1:0.5(w/v). The result showed that the content of piperine was 77.14%, the extractive yield attained 83.96%.Compared with the technology of water distillation, SFE-CO2 of pepper oil was proved to be efficient and save time. Meanwhile the extractor of SFE-CO2 has sweet-scented and remains the smell of original crude materials. Compared with the technology of traditional solvent extraction, SFE-CO2 of piperine was proved to be simple and efficient. The extractor of SFE-CO2 has light color and smell peppery.The produce cost of SFE is lower than that of alcohol solvent extraction by preliminary economic accounting. SFE-CO2 is feasible to extract Piper nigrum Linn. So the craft of the SFE-CO2 of Piper nigrum Linn is applicable.By using crystal for isolation of the extractor of SFE-CO2, the solubilities of piperine in Ethanol, ethyl acetate and aether were determined respectively. By using the content of piperine in crystal and the extractive yeild of peperine as quality control criteria, The effects of solvent, crystal temperature, rate and time on the crystal results of Piper nigrum Linn were explored . The crystal conditions for piperine were as followed: solvent temperature 40℃, descent rate 1℃/5min, crystal time 4h, the ratio of the extractor and solvent was 1:4-1:5(w/v). The result showed that content of piperine was about 90%, the extractive yeild was 60~70%.
