Urease is an effective enzyme which specially catalyze hydrolyze of urea, it's difficult to keep the activity of free urease . Immobilization of urease can solve the problem, it have a good stability, and furthermore it can use repeatedly.This text did the research of the part to the physics and chemistry property of terrabacter tunescens and immobilized urease. The main concentration is taking the Sodium Alginate and the Polyvinyl alcohol as the body to immobilize urease, in the meantime carrying on a research of immobilized and free urease at the best respond temperature,the best responded pH,stability...etc. The Terrabacter tumescens is high to produce the Urease,the development temperature is 30 ℃, the most suitable respond temperature is 55 ℃,It can afford higher temperature . The germ's stub absolute aerobic to develop, the most suitable is pH 7.50, The most value of producting Urease is in the pH 7.5. The adoption of urea to induce the Urease can reach to 300% of not. Shaking strength can also raise the yield of Urease effectively .Two kinds of material packs cover up the fixed microorganism urease can make urease of the heat-proof temperature raise 10 ℃, 55 ℃ raise to 65 ℃ from the most suitable temperature. The immobilized urease vitality comparing to free urease with respond the pH variety curve become flatter . The enunciation turns the pH scope more breadth of the immobilized urease to free urease . The immobilized microorganism urease have a storage stability fixedly.The storage stability of the cell fixedly is obviously high to leaving cell under 0-4 ℃. Using Polyvinyl alcohol to turn cell fixedly is slightly high than Sodium Alginate in the vitality recovery and the reaction stability.
