| Tobacco is an economic crop for its leaves and needed for its taste sensation. The quality of tobacco related products is determined by a multitudinous variety of substances in tobacco leaves, which are extremely complex mixtures. Compositions of different tobaccos are not the same.In the thesis, high performance liquid chromatographic (HPLC) methods were developed for analysis of amino acids, carotenoids in tobacco. Amino acids and carotenoids were analyzed by solvent extraction HPLC. Dalian Yilite ODS C18 reserved-phase column (3 μm, 100×4.6 mm) is used for the separation of amino acids and carotenoids of the tobacco. Amino acids were separated by linear gradient elution using water-acetonitrile (1:1, V/V) and sodium acetate-acetic acid buffer solution (pH 6.4; containing 10% acetonitrile, V/V) as mobile phase; carotenoids were separated by linear gradient elution using ethyl acetate and water-acetonitrile (1:9, V/V) as mobile phase.Two kinds of compounds in tobacco mentioned above were qualitatively analyzed by RT (retention time), RRT (relative retention time), spectrum and standard sample adding and quantitatively analyzed by internal standard. The simple pretreatments provided good RSD (amino acids: 2.2-5.0%; caroenoids: 2.6-6.0%) and high recovery rate (amino acids: 85.3-101.2%; carotenoids: 92.7-102.1%).Based upon the research above, we relatively determined the quantity of amino acids, carotenoids in about 100 different tobacco leaves collected by Changde Cigarette Company and compared their mutual proportions.HPLC fingerprints were developed, for identification of the complex components of the casing for tobacco. The samples were separated with a Waters symmetry C18 reserved-phase column (5 μm, 250×4.6 mm) by linear gradient elution using water-formic acid (1000:1, V/V) and acetonitrile as mobile phase. Mean chromatograms and correlation coefficients of samples were calculated by the software "Similarity Evaluation System for Chromatographic Fingerprint of TCM".Results of this thesis provided data for intelligentized cigarette design and helped to reduce tar and speed the development of cigarette industry. |
