The pharmacological function and the development of central stimulant Caffeine have been described in this paper.Two synthetic routes of Caffeine have been determined and investigated after the analyzing and trying for the literal synthetic methods. In one route, the target compound was prepared from chloroacetyl chloride through six steps, the total yield is 50.5 %. In another route, the target compound was obtained from cyanoacetic acid through six steps, the total yield is 45%. The new designed synthetic routes resulted in decreasing the cost, simplifying the operation procedure, and was more suitable to industrialization. The structure of Caffeine had been confirmed by ~1H-NMR and MS.By the experimental uniform design, the synthetic processes of synthetic intermediates such as 6-imino-1,3-dimethyl-dihydropyrimidine-2,4-dione and 1,3-dimethyl-4-amino-5-nitrosouracil were optimized. The best reaction conditions had been determined after the investigation for the factors of reaction, such as ratio, reaction time and reaction temperature.
