Abstract: Four high active soybean isoflavone glucoside hydrolase producing mould strain, sp.R, sp.42, sp.00 å'Œ11 sp.48,ware selected from sp.R, sp.S, sp.39, sp.42, sp.00, sp.48. sp.848 å'Œ sp.9. The result showed that sp.R was the highest active soybean isoflavone glucoside hydrolase producing mould strain by liquid fermentation. Its optimal hydrolase-producing conditions were as follows: 2.5% wheat bran as carbon source, 1% NaNO3 as nitrogen source, initial pH7.0, culture medium volume 40mL/250mL, inoculating quantity 8%,culture temperature 30"C, revolutions 160r/minand culture 84h. The enzyme activity reached 82U/ml.Study the physiology activity of the soybean isoflavone. Through the method that three phenol of neighbor benzene from oxidize and ratio the color method measurement the Fenton responds the method of the output Hydroxyl free radical, oxidizing the activity to carry on to the anti-of the soybean isoflavone comparison research.through the MTT method. The result showed the enzyme solves glucoside yuan, Soybean glucoside to the dosage that clearance rate need when attain 50% of the super oxygen free radicals namely the IC50 distinguishes for the 0.14 mgs/ tnL, 0.32 mgs/ mL, to the dosage that clearance rate need when attain 50% of I lydroxyl free radical namely the IC50 distinguishes for the 0.57 mgs/ mL, 1.04 mgs/mL.The soybean isoflavone represses to the big rat cancer of the liver Cell's system CBRH-7919 growth the rate attains 50% need of dosage namely the IC50 is a 7.55 mgs/ L, repressing to the small ratleukemia CML- K562 Cell's system growth the rate attains 50% need of dosage namely the IC50 is a 10.35 mgs/ L. The antioxidant properties of the aglycons hydrolysis by enzyme is better than the others.
