Behavior of the nattokinase which is very beneficial for health,and the active mass of soybean—soybean Isofiavone and Soyasaponion,in the fermentation of Natto strain(Nat to Bacillus 918)was examined.And optimize the condition of ferment.The soybean isoflavone was extracted by alcohol.The most of extraction from soyabean by alchol was genistin.The pure genistin and acetygenistin were obtained by the Sepabeads SP207 macro-pore resin.The Soyasaponion was extracted by alcohol and n-butanol,the pure soyasaponion was obtained by the AB-8 macro-pore resin.Determin the extraction method of nattokinase,and choose the more convenient and rapidly assay method to test the nattokinase activity—Bacillus Subtilis proteinase activity.Avoiding the complex method which to detect the UK,degrade the difficulty and expense.The optimal extract condition of the soybean isoflavone was,80%alchol,70? extracted 3h by the dosage liquor of 1:6;The optimal extract condition of the soyasaponion was,70%alchol,under the room temperature extracted overnight by the dosage liquor of 1:6.Behavior of the nattokinase which is very beneficial for health,and the active mass of soybean—soybean Isofiavone and Soyasaponion,in the fermentation of Natto strain was examined.In the Natto solid state fermentation,the nattokinase production was higher and the glycoside convert to aglycon,in the condition which water content was 80%,the pH was 8.0 at 40? cultured for 48h..?According to the HPLC method,it can appear obviously,the soybean Isofiavone,which was hydrolyzed by enzyme convert to the aglycon.It can improve the activity utility.
