Rheumatism is a disease which is caused by wind, cold, dampness, hot invasion body, with the symptom of ache and numbness of extremities bones and muscles, the tumefaction and calor of arthron, or activity disabled. The reason of illness is not completely clear, but is related to immunodeficiency, inheritance and infection. Recently, the traditional herb medicine get rapid development in the therapy of rheumatism, especially the herb medicine preparation for external use. But, the traditional preparation and the extraction technology restricted the development of the preparation of the herb medicine. The traditional preparations of the herb medicine were black drug plaster and adhesive plaster, which content of effective component were more lower and the dosage were low. The wanlingwuxiang cataplasma studied in this articles not only maintained the certain effect of the traditional drug plaster, but also exert advantages of comfortable, ventilated, wetness, no irritation to skin, etc, which have better market prospect than the two dosage forms before.This article studied the extraction technology of the effective content in wanlingwuxiang cataplasma, and optimized the extraction technology of extracting volatile oil that have the effect of activating vital energy circulation, alleviating pain, promoting blood circulation to remove blood stasis using orthogonal experimental method. The article also studied the factor which have influence on the content of cinnamaldehyde and volatile oil in extract, this factor include extraction temperature, extraction pressure, the flow rate of CO2, the size of the medicine particles, separating temperature, extraction time, etc. At last, we screened a better extraction condition, which extraction pressure was 25Mpa, temperature was 50℃, he flow rate of CO2 was 30kg/h, separating temperature was 40℃, extraction time was 1h which could get the best extraction effect. With the mark of the content of liposoluble alkaloid and strychnine, using ethanol reflux method, we optimized the technology which extracted the alkaloid with the effect of expelling wind and dampness, anti-inflammation and alleviating pain. The best extraction condition was that the size of the medicine particles was the biggest, using 7 times 90%ethanol, extracting 3 times and extracting 1.5h every time.This thesis established a range of property item of cataplasma, confirmed the detecting mark of tackness, adhesive, cohesive, tolerance, rubber strength, the rate of water loss, and controlled the inner physical quality of cataplasma basically. We studied several factor including different matrix materials, the filling agent, adhesive agent, wetness agent, adjustment agent, with the quality mark of adhesive and physical character of the cataplasma, and every factor and dose were optimized using orthogonal experimental method. The result showed that the base with the proportion of glycerin: PANA: gelatin: PAA: CMC-Na: Sorbitol: kaolin = 2.5: 1: 0.3: 0.2: 0.6: 0.3 was the best. We carried out a range of inspection for the relating factors in preparating technology of cataplasma, and inspected the quality of preparating technology after optimization.To study the effect of various new type penetration enhancers on the percutaneous permeability of muscone in wanlingwuxiang ctataplasma in vitro.Thewanlingwuxiang ctataplasma containing various penetration enhancers such as Transcutol p, Labrasol, Labrafil 1944, Azone , propylene glycol, N-methyl-2-pyrrolidone were preparated. The steady state flux of muscone from ctataplasma were determined in vitro on exciced mouse skin using modified Franz diffusion cells. The accumulation permeation amounts and permeation rate were calculated. The result showed that the diffusion behavier which muscone penetrated mice skin was coincided with Fick's diffusion equation, and Q was in line with T. Transcutol p, Azone and N-methyl-2-pyrrolidone showed significicant enhancement effect on the permeability of muscone. The maximum enhancement was obtained in wanlingwuxiang ctataplasma with Azone/ propylene glycol /Labrasol(3:15:6).The LC methods for the assay of muscone in cataplasma were established, the assay methods of LC programmed thermogence were confirmed. The linear relation , the precision experiment, the stability experiment, the repetition experiment, the recovery rate experiment were investigated. Result of the methodology showed that the confirmed method was stabled, having good repetition and recovery rate which reached 101.4%. We also investigated the assay of chuan-wu and nux vomica, and the content of aconitine and strychnine were limited by thin layer chromatogram method.The initial pharmacodynamics study was carried out for the prepared cataplasma. We investigated the effect of anti-inflammation and expelling wind and dampness, by anti-mouse adjuvant arthritis experiment and anti-mice croton oil ear swelling experiment. We also investigated the effect of alleviating pain, by mice light-hot method and mice twist response experiment. The result showed that this preparation had obviously effect in anti-inflammation and alleviating pain.Acute toxicity experiment and local irritation experiment for skin were carried out in toxicology studies. Obviously toxicity response in acute toxicity experiment was not observed when the dosage of the cataplasma was 20.0g/kg (which was 400 times than the dosage for human). The preparation exert no irritation not only for integrated skin, but also for damaged skin.
