Urease is an effective enzyme which specially catalyze hydrolyze of urea, it's difficult to keep the activity of free urease. Immobilization of urease can solve the problem, it have a good stability, and furthermore it can use repeatedly.In this paper, I have do a partial research in the immobilization of urease. Concentrate on glutin and cellulose acetate as carrier to immobilization and cross-linked urease congeries to immobilization, which have no carrier. Use film of glutin as carrier to immobilize urease, the activity of immobilized enzyme is 3.140U/g, and the yield activity of enzyme can attain to 49. 1%, even have more yield activity if we increase the dosage of free urease. Use film of cellulose acetate as carrier to immobilize urease, the activity of immobilized enzyme can achieve 10. 699U/g easily, and the maximal yield activity of enzyme can attain to 37. 16%, The activity of cross-linked urease congeries can reach 22. 430U/ml, and the maximal yield activity of enzyme can reach 31.80%. We also have research the km and the reaction condition and the stability of free and immobilized urease.
