Nine highly active cellulolytic strains, one bacteria and eight fungi, were isolated from Zijin Mountain in Nanjing. The cellulase activity of four fungi were higher than Trichoderma reesi 3.3010 and Trichoderma viride 3.3711, which were conserved in our laboratory and bought from CGMCC, whose cellulase activity were acknowledged high. Then one bacteria and one fungus were identified. The results showed that the bacteria is Cellulomonas, named LBT2-8 and the fungus is Trichoderma, named MJ1. The degradation ratio of LBT2-8,MJ1 and Trichoderma viride3.3711 were determined.Then the extracting method of crude enzymes was studied. By flask solid-state fermentation, the effects of different mediums and fermentation conditions on cellulase production by LBT2-8 and MJ1 were studied. Subsequently enzymatic properties of the crude enzyme filtration from LBT2-8 and MJ1 were investigated and the optimal temperature and pH for the enzyme reaction were studied. At last protein content of MJ1 fermentation, MJ1 and yeast co-fermentation were determined. |
