Ascidian , a kind of marine organisms , has received significant attention for its role in ecology restoring and abundant active compounds in its body in recent years. Research on seperation and pharmaceutical application has been undertaken on.The present study will focus on the determination of process parameters for extracting fatty oil from ascidian and improvement of oxidative stability for corresponding fatty oil.Based on a series of experimental work, it is found that ethyl acetate can be used as the most suitable organic solvent to extract fatty oil from ascidian . the optimized conditions of organic solvent extraction determined by orthogonal experiment are as follows: temperatue 40℃, extraction time 3h, w(weight of the powder of ascidian):v(volume of ethyl acetate ) 1:4 .The yield of fatty oil is approximately 1.80% under this condition.The fatty oil extracted was analyzed by gas chromatography. Analytical report shows the relative content of polyunsaturated fatty acid in the total fatty oil is 67.70%, and the sum of EPA and DHA is 21.81 %.According to the results of single factor test and orthogonal experiment, the optimization conditions of SCF-CO2 extraction are as follows: the granularity of the powder 40~60 grade, temperature 35℃, pressure 25MPa, volume of carrier entrainer(ethyl acetate) 100ml, time 2h and the flow rate of CO2151/h. The yield of fatty oil is about 1.95% under the extraction condition. The fatty oil extracted was analyzed by gas chromatography. Analytical report shows the content of polyunsaturated fatty acid in the total fatty oil is 73.31%, and the sum of EPA and DHA is 33.96%.Because fatty oil extracted from ascidian contains a relative high concentration of polyunsaturated fatty acid and can be oxidized easily, it is necessary to add appropriate antioxidant in the fatty oil to prevent it from being oxidized.The experiment results show inducement phase of the fatty oil extracted by the way of SCF-CO2 is increased to 63.8h when Ve with the concentration of 2.0×10-4g/g is added, and the inducement phase of the fatty oil extracted by the way of organic solvent is increased to 58.3h when the extract of salvia plebeia with the concentration of 2.0×10-4g/g is added. Therefore two kinds of fatty oil extracted by different ways both reach a set standard of storage.It is possible to develop the fatty oil from ascidian as health product depending on the above process and improvement of oxidative stability.
