In this research, by the single facter optimization and the orthogonal test , the optimum submerged culture medium and culture conditions of Torula utilis for the reduced glutathione (GSH) were studied. The results of the variance analysis showed that the composition of the optimum medium was glucose 1.5%, peptone 1.0%, Cysteine 0.15%, Glutamic acid 0.05% Glycine 0.05%, MgSO4 0.125%, and the optimum suberged culture temperature , initial pH, medium capacity of the shake flask and inoculation rate were 28℃, 6.5, 50mL/250mL and 10% respectively. When the inoculation rate was 10%,the biomass (dry) and the yield of GSH were comparatively high. The concentration of glucose-added effected on the yield of GSH greatly:from the fourth hour,2h interval, three times,the conc. of glucose was added 5g/L every time. If the conc. of the total glucose was beyond 30g/L,the yield of intrcellular GSH was low. When the conc. of the total glucose was 25g/L, 30℃, 120r/min, 24h,the biomass and the yield of intrcellular GSH were 8.58g/L and 13.55mg/g respectively,the yield of GSH in the fermented broth was 118.5mg/L。The total GSH was 2.656% of the biomassBy the single facter optimization and the orthogonal test , the optimum submerged culture medium and culture conditions of Morchella esculenta for intracellular and extracellular polysaccharide were studied. The results of the variance analysis showed that the composition of the optimum medium was sucrose 2.0%, Corn extract 2.0%, Soybean extract 1.0%, Urea 0.5%, KH2P04 0.05%,MgSO4 0.05%,FeSO4 0.1%,ZnCl2 0.1%, and the optimum suberged culture tempertrature , initial pH, medium capacity of the shake flask , inoculation rate and fermented time were 22℃, 7.0, 100mL/250mL 10% and 5d respectively. The yield of intracellular polysaccharide was3.14g/L and the yield of extracellular polysaccharide was 0.78g/L 17.0g/L dry weight of Morchella esculenta mycelia was gained.At the basis of modern nutrition theroy, a formular for products was studied.
