With the developmen of molecule biology, the function of polysaccharide and its compound were further studied. G.lucidum polysaccharide(LPS) and T.matsutake polysaccharide(SPS) is one of the main functional ingredients in G.lucidum and T.mastutake.Modern researches show that:LPS has many active functions:regulate immunity,antitumor,promote metabolism of nucleic acid and protein, antioxidant, radioresistance, antiinflammation,lower blood sugar,and so on;SPS also possion many functions, such as:strong and healthy, good for intestines and stomach,ease of pain,reduce phlegm,tetramisole,cure diabetes,antitumor and so on.Polysaccharide from fruiting body cultured has both splendid for treatment and dis advantages such as: influence of season and weather,long period,lower productivity, high cost.Submerge culture are paid more attention.Research of co-fermentation,however,is rarely reported.This paper is studied about fermentation characteristic,extration, isolation, purification,identification and antitumor of polysaccharide of G.lucidum, T.matsutake and L-S co-fermentation (mutagensis and well-seiection).The biological and kinetic characteristics of G.lucidum,T.mastutake,L-S Co-fermentation in shake flask and in 10L fermentor.The results showed:After 5d,the biomass,exopolysaccharide,endopolysaccharide and total polysaccharide of G.lucidum were 11.21g/L,0.89g/L,1.17g/L,2.06g/L respectively in shake flask and 10.11g/L ,0.88g/L ,1.048g/L, 1.928g/L separately in 10L fermentor;3d later, the biomass,exopolysaccharide,endopolysaccharide and total polysaccharide of T.mastutake were 10.4g/L,0.15g/L,0.442g/L,0.592g/L respectively in shake flask and 11.51g/L, 0.26g/L,0.437g/L,0.697g/L separately in 10L fermentor;The biomass,exopolysaccharide,endopolysaccharide and total polysaccharide of L-S Co-fermentation were 11.43g/L ,0.751g/L,1.09g/L,1.841g/L respectively in shake flask and 10.65g/L ,0.858g/L,0.909g/L,1.767g/L separately in 10L fermentor for 4d.The optimum culture conditions for L-S co-fermentation was studied by orthogonal test.The biomass and exopolysaccharide of L-S co-fermentation were up to 11.43g/L, 0.751g/L separately under the optimum submerged culture condition,such as: submerged culture temperature,initial pH,medium capacity of the shake flask and the rotation rates were 28oC,PH 5, 80ml/250ml, 130r/min respectively.The extraction process of endopolysaccharides from G.lucidum,T.mastutake,L-S Co-fermentation was optimized by orthogonal test.The results show that :the maximum yields of endopolysaccharides from G.lucidum,T.mastutake,L-S Co-fermentation were up to 10.37%,3.80%,8.54% respectively under the following conditions:extraction temperature 95 oC; Rate of fruitbody power to water1:20; Extraction time2h.The extraction factors of concentratio of alcohol and extraction times were determined by single factor test. Concentratio of alcohol and extraction times were 75-80% and twice respectively.G.lucidum ,T.matsutake and L-S co-fermentation extracellular polysaccharidewere purifiedby the procedure including Savage method,DEAE-cellulose column chromatograph and Sephadex G-100 gel filtration.The structure of the extracellular polysaccharides purified have been determinated by the methods of GC and Sephadex G-75 gel filtration.The comparison of the effect of G.lucidum Exopolysaccharide,T.mastutake Exopolysaccharide,L-S co-fermentation Exopolysaccharide and L+S mixed Exopolysaccharide on the anti-tumor of mice was studied.The results showed that: the optimum inhibition rate of mice reached 85.6% in 450mg/kg.d of G.lucidun exopolysaccharide,88.1% in 150mg/kg.d of T.mastutake exopolysaccharide.The effect of L-S co-fermentation exopolysaccharide on the inhibition of mice was the best,which could get to 89.2%;The optimum inhibition rate of L+S mixed exopolysaccharide was not so good as that of L or S exopolysaccharide alone.It was only 77% in 150mg/kg.d dosage.
