Application of Photo-Chemical Analysis (UV and Fluorimetry) in the determination of pharmaceuticalsAbstractsThis presentation includes the following parts, first, determination of urapidil by UV and fluorimetry, second, analysis of aspirin and salicylic acid by synchronous fluorimetry and third, determination of dissolution of 5-FU by FI-UVA and the last study on chitosan-immobilized urease and its application.Part 1: Review.A somewhat thorough and all-around review of the application of fluorimetry in pharmaceutical is given in this part.Part 2: Determination of aspirin (AS) and salicylic acid (S) by synchronous fluorimetry.I Simultaneous determination of AS and S by synchronous fluorescence spectrometry.A simple, rapid and inexpensive fluorimetric method for simultaneously determining AS and S in a single scan of the co-existing specimen is described. The results demonstrate that the average recoveries of AS and S are 98.5% and 99.5%, RSD are 7.1X10'5 and 1.6X10'4%, respectively. The limit of detection is 4.0X10" 7mol ?L"1 for S. Satisfactory results were obtained by this method.II Determination of aspirin and micro-amount salicylic acid in pharmaceuticals by synchronous fluorimetry.A synchronous fluorimetric method is described for determining AS and S in pharmaceutical specimens, with A A =50nm for AS and 130nm for S. The average recoveries are 100.3% and 98.6%, and RSD are 2.6% and 11.32% for AS and S, respectively. The results show that it is a satisfactory method in the determination ofAS and S.Part 3: Determination of urapidil by fluorimetry and UV spectrophotometry.I Study of the fluorescence spectrum of urapidil and its determination.A simple and rapid method of determining urapidil by fluorescence spectrometry is described. Urapidil will give out a considerable large of fluorescence strength when irradiated by a UV light and thus a new way for determination of urapidil is established. The linear range is 3.30X 10'7-1.65X lO^mol -I/1, R2=0.9992, and the mean recovery is 98.99% for tablets and 99.80% for injections .The relative standard deviation is less than 1.0%. Satisfactory results were obtained by this method.II Study of the UV spectrum of urapidil and its determination.The UV absorption characteristic of urapidil is studied and another method of determining urapidil by UV spectrophotometry is reported. The linear range is 9.91 X 10'7-4.96X 10'5mol-L'' and the mean recoveries for tablets and injections are 99.65% and 99.85%, respectively. The relative standard deviation is less than 1.0%. It shows to be a rapid and simple method in the determination of urapidil without any premier chemical reaction or solution pretreatment and can be an alternative of pharmacopia.Part 4: Flow-injection UV spectrophotometric study on the 5-fluorouracil release rate in chitosan-alginate microspheres.The chitosan-alginate microspheres serving as carrier for 5-FU were made and the drug release rate was studied by flow-injection continuous sample-induced and interval sample-induced spectrophotometric method respectively. The effect of different concentrations of chitosan and alginate in the preparing process was investigated. The results were discussed and the.two methods were compared.Part 5: Study on chitosan-immobilized urease and its application.This part deals with: (l)the conditions of the preparation of chitosan microspheres, (2)some characteristics of chitosan-immobilized urease, and(3)theIVexample of the usage of the chitosan-immobilized urease as a chemical sensor for the determination of urea in waste water. The linear range is l.OX 10"5-5.0X lO^g-and the mean recovery is 100.5%.
