Human milk is the best food of baby, more than 98% of the lipid in the human milk is triglycerides,It is one of the main nutrient components of human milk,It’s not only can provide baby part of the energy required for growth,and can provide the must fatty acids for baby growth,and it is very important to the growth of brain organ.The fat in human milk have special structure features, among this part of fatty acids,45%~55% is saturated fatty acids,most of saturated fatty acids is palmitic acid (quality content is 20%~25%),And more than 70% of palmitic acid was distributed in Sn-2;15%~20% fatty acid in Human milk fat was not more saturated fatty acid,the most part of unsaturated fatty acid is oleic acid,mainly distributed in Sn-1,3.There are two commonly used methods among the preparation methods for substitutes of human milk fat.One is using several or many kinds of oils in specific lipase catalytic effect of ester exchange reaction.The other method is various fatty acids and oils having a (acid solution) ester exchange with 1,3 specific lipase catalyzed transesterification reaction.In this study,the main triglyceride composition of triglyceride is 1,3-2 oleic acid-2-palmitic acid triglycerides (OPO).Using a two-step reaction to synthetize OPO:The first step,three palmitic acid triglycerides (PPP) preparation,glycerol and palmitic acid were reacted in high temperature with sulfuric acid;The second step,ppp and oleic acid were reacted with 1,3 specificity lipase biological catalyst to synthetize OPO.In the purification of the product,we compared three methods for separating fatty acid.Vacuum distillation molecular distillation and Alkali refining can separate the fatty acid in product.Then we optimized the parameters in this method for incressing the separation effect.In the first step reaction,We discussed the effect of temperature,molar ratio of the production rate of PPP;In the second step reaction we dicussed the influence of temperature,molar ratio,lipase and rotational speed in the conversion rate of OPO.The technology of this paper had carried on pilotscale experiment,The results of this pilotscale experiment shows that the technology has good stability.account the producing cost of the pilotscale experiment,the result prove that this process has economic feasibility.
