Recently micro / nano nanometer materials has a lot of synthetic methods, commonly synthetic methods including sol-gel method, hydrothermal method and hard template method, soft template method, sacrifice template method, etc.; Inorganic nanometer materials is widely used in fluorescent tags, drug delivery, sewage treatment, lithium batteries, catalysis, sensing, and chemical energy storage, etc. micro / nano materials calcium phosphate and calcium carbonate have been widely used in loading, drug transport and controlled release field because of their good biocompatibility, biodegradability and pH sensitivity. Surfactant mainly through soft template, adsorbed on the surface of nanomaterials changepolymorphs growthorForm complex ion with synthetic materials tocontrol the morphology of product; Biomimetic synthesis is based on the basic principles of biomineralization, simulation of biological processes in vivo formation of biominerals, the use of biological molecules in vitro regulation of the synthesis of inorganic materials.Compared to conventional drug treatment, chemotherapy and radiation therapy,the presence of drug delivery system(DDS) has the following advantages:high concentrations tumor lesion sites, drug side effects, and low frequency of administration; Targeted drug delivery system(TDDS) includesthree reagents:carriers, targeting and drugs, has four characteristics: keeping, avoiding, targeting and releasing. Before reaching the tumor site,the marerialscan encapsulated in the interior of the drug, it is possible to avoid the phagocytosis of monocytes, can be targeted to the tumor site identification, release of the drug, can be divided into passive targetingand active targetingaccording to the carrier mechanism and site of the lesions; Passive targeting is the natural target, active targeting is modify on the surface of the drug carrier to connected with the specific receptor ligandson the surface of tumor cells. In the process of drug delivery,the ligandon the surface of carrier material and receptor on the surface of the tumor cells bonded specifically, to achieve targeted delivery effect. Compared with the micelles, organic composites, liposomal carrier,calcium phosphate and calcium carbonate drug system embodied in the non-toxic, stable and good biocompatibility, good pH sensitivity and easy to discharge; using the tissue around the tumor cells and the intracellular pH value of the lysosomes were lower than normal cells of pH value so as to achieve the purpose of targeted drug release.This thesis is divided into the following areas:FA and CTAB for organic matrix, we synthesized two flower CaCO3 materials, UFO-like CaCO3, vesicular phosphorus Ca3(PO4) 2, spherical and dandelion-like BaCO3, the morphology of the sample and properties were characterizedthrough a variety of different means of testing, the results show that the morphology of materials unity, having a porous structure, and the surface potential is negative, these properties help to improve the load rate of the material.With DOX as a model of anticancer drugs, we conducted drug load-release experiments in vitro with CaCO3 and Ca3(PO4) 2 materials,the test results show that the flower-like CaCO3, FA-CaCO3, UFO-like CaCO3 and Ca3(PO4) 2 have highdrug loading rate, respectively, 95.9%, 84.5%, 83% and 82.5%, Controlled release tests showed that: due to calcium carbonate and calcium phosphate materials are pH-sensitive materials, In weakly acidic conditions can be decomposed slowly to release the drug, does not decompose under neutral conditions, protect drugs and reduce drug release in the surrounding normal cells, and in former 50 h there’s no explosive release phenomenon, can achieve the effect of slow release, a slow controlled release of drug release time can be extended, reducing the frequency of administration.This result provides important experimental evidence for cell viability test material.Cell biology test results show that drug specificity is obvious when the DOX load in the two flower-shape CaCO3 materials, especially the specificity of the FA- CaCO3 material can be increase to 3.3 times that of the pure medicine and the UFO-shape CaCO3 material can be increase to 3.3 times that of the pure medicine.The result shows the synthesis of FA- CaCO3 and flying saucer shape CaCO3 in the field of anti-tumor drug load and tumor therapy has good potential applications.
