| Miaoyu Dangshen belongs to Chuan Codonopsis, Its main origin is Chongqing Wushan, Daping (Duping) and Miaoyu county and the name due to the Dangshen distribution center on Miaoyu county long time ago. Miaoyu Dangshen has been ranked the first of "China’s four dangshen" once in a while. But in recent years, many lacks of the basic research and industry chain on it, which turns out a single Dangshen product and low value-added products, Miaoyu Dangshen need to develope in stimulate now. Therefore, this paper commissioned by Wushan government and Shennv pharmaceutical, funded by the Chongqing University Cooperation Projects (No.104290-41003012), it would be hoped to achieve more economic value through the vigorous researches on Miaoyu Dangshen.Objective:This compound health beverage is made up of Miaoyu Dangshen, Polygonatum, wolfberry, and Chrysanthemum, the paper aims to build a complete preparation route and formulate a quality standard for Dangshen compound health beverage (DC). Its health effects on anti-fatigue, anti-radiation will be designed to research.Methods:1. The preparation of DC health beverage: With the extraction yield, and the contents of chlorogenic acid,3,5-O-Dicaffeylquinic acid as indicators, the heat reflux extraction technology was optimized by orthogonal design. After the optimization, the extract was purified by chitosan, the purification technology was optimized with the orthogonal design and the contents of polysaccharide, chlorogenic acid,3,5-O-Dicaffeylquinic acid as an index to select the best purification process. Finally, an appropriate amount of flavoring agent will be added to it, and the initial quality standards of it will be established.2. Anti-fatigue function evaluation:According to the evaluation procedure and test method of functions of healthcare food of the Ministry of Health in China, we evaluated the exhaustion swimming time, the glycogen content and the serum urea nitrogen of mice after mice administered the product 30 days.3. Anti-radiation function evaluation: After mice administered this product 30 days, establishing the radiation model in mice. Then, at different time points, detecting the survival rate of mice, peripheral blood compound, organ index, bone marrow nucleated cells, bone marrow cell cycle and apoptosis of mice.4. Safety assessment:The largest one-time oral administration tested in mice, the maximum dose of the prescription on mice be recorded. After mice administered this product 30 days, the liver and kidney were taken hematoxylin-eosin staining to observed morphology changes.Result:1. The preparation technology of DC health beverageThe 4 kinds of medical materials were taken based on the prescription. The materials were mixed in 2.19 times amount of water (g/mL) to make up the amount of water absorption, besides, add a 12 times amount of water (g/mL) to soak. After 40 minutes, the mixture was reflux extracted for 2 times,1 hour for each, then the extract was obtained after filtration. The filtrate was condensed to 0.25 g/mL, after incubated at room temperature, added with chitosan solution to make the chitosan solution content achieved at 20%, placed it in the 50℃ water bath, removed it after 10 minutes. After setting for 24 h at room temperature, the mixture was filtrated, the 16% filtrate and 1.44% white sugar were mixed with water to prepare a prototype, through the fine filter, potting, sterilization 25-30min at 90-95℃, the Dangshen compound health beverage was successfully obtained.2. Anti-fatigue function evaluationAfter these mice administered this product 30 days, we observed that this product can significantly prolonged the exhaustion swimming time of mice, increased their glycogen, reduced the amount of serum BUN (p<0.01).3. Anti-radiation function evaluationThis product had a significant effect on spleen index, bone marrow cell cycle, when compared with the radiation control group (p<0.05). it can be significantly reduced the radiation damage in mouse spleen, promoted the proliferation of bone marrow cells.4. Safety evaluationThe maximum oral dose of the prescription on mice is greater than 5.71g (crude drug)/kg, there is no significant toxicity in mice. After these mice administered this product 30 days, we observed these liver and kidney tissue had no significant pathological changes.Conclusions:This product had a clear anti-fatigue effect and anti-radiation effect. A complete and reliable DC health beverage process route have be formulated and the initially quality standards need to established. The radiation protective effect and mechanism of hematopoietic system in mice need further study. |
