In order to study the new chelators, which can decorporation the neclear metal and repair the body simultaneously, three novel fullerene pyridinone derivatives were synthesis, and the capability of complexation with uranyl ion,scavenging the free radical, including the cotoxicity were researched.(1)Starting from the maltol, the hydroxyl were protected with benzyl bromide and proceeded to produced 3-benzyloxyl-2-methyl-pyrone, then reacted with malonyl dichloride to yield the five kinds new malonamide 3-benzyloxyl-2-methyl-pyrone. To improve the water-solubility of the target compounds,removal the benzyl from the oxygen to obtain the five new tetradentate chelators(tetradentate HOPO) at first step, then were introduced into the fullerene with the Bingel reaction. Three novel water-soluble meathol fullerene pyridinones multiaddition derivatives(C60-HOPO), which most was trisaddition derivatives were obtained, and all the structure of the products were characterized by FT-IR、Uv-Vis、1H NMR、13C NMR and MS.(2)The chelating ablitity of tetradentate HOPO towards the uranyl ion were studyed under the condition of p H at 7.4 on method of equivalent molarity. While,the C60-HOPO were studied under various range of p H on the method of SCP competed chelating. What’s more, the selectivity of the C60-HOPO toward normalized metal ions in human body, such as K+、Ca2+、Mg2+、Zn2+, were studied with the fluorescence quenching methold, the results indicated that the C60-HOPO/UO22+ presented a good stability without any significant complexation with the normalized metal ions.(3)The DPPH free radical scavenging of C60-HOPO was researched and it show a effective capability. Addition, the superoxide radical anion scavenging of C60-HOPO were studied in autoxidation for pyrogallol system and the scavenging capability was more 100 times significant than vitamin E, it is an effective radical scavenger.(4)Therefore cytotoxicity of C60-HOPO were comprehensively evaluated using two different cell lines(A549 and L929) on the method of MTT and ffluorescence sign counting, the results indicated that the C60-HOPO show evidencecytotoxicity as expected.
