Study On Fullerene Derivative Drug Delivery System | | Posted on:2021-01-28 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Wang | Full Text:PDF | | GTID:2381330602971522 | Subject:Chemical engineering | | Abstract/Summary: | PDF Full Text Request | | In this paper,three tumor drug delivery systems based on fullerene derivatives loaded with anti-tumor drug doxorubicin were constructed:p H-sensitive tumor drug delivery system(FP-DOX)and folic acid-loaded tumor drug delivery system(FP-FA/DOX)and ferrous oxide loaded tumor drug delivery system(FP-IONP-COOH/DOX),which also investigated the drug release rule and free radical scavenging ability,the main research contents were as follows:1.Researched on p H-sensitive tumor drug delivery system(FP-DOX)based on carboxypropyl fullerene pyrrolidine drug delivery system:FP-COOH was used as a drug carrier,and hydrazone linkage was connected to DOX to form a p H-sensitive tumor Drug system(FP-DOX),in which UV-vis,FT-IR and MALDI-TOF respectively verified the effective synthesis of FP-DOX,solubility experiment showed that FP-DOX could effectively improve the water solubility of fullerene.The in vitro drug release experiment indicated that FP-DOX was a p H-sensitive preparation.2.Researched on a folic acid-loaded tumor drug delivery system(FP-FA/DOX)based on a fullerene monoaddition pyrrolidine drug delivery system:the target molecule folic acid was connected to FP-OH through esterification reaction loaded with doxorubicin,constituted a tumor drug delivery system(FP-FA/DOX).Among them,UV-Vis,FT-IR and MALDI-TOF respectively verified the effective synthesis and preparation of FP-FA/DOX,and the in vitro drug release experiment showed that FP-FA/DOX had a slow-release DOX release rule.3.Researched on tumor drug delivery system(FP-IONP-COOH/DOX)loaded with ferrous tetroxide based on carboxypropyl fullerene pyrrolidine drug delivery system:using hydrothermal chemical bonding ferric oxide was loaded on FP-COOH,and DOX was physically loaded to form a tumor drug delivery system(FP-IONP-COOH/DOX).Among them,UV-Vis and FT-IR verified the effective synthesis and preparation of the product,respectively.The determination of the curve showed that FP-IONP-COOH had superparamagnetic properties,and the in vitro drug release experiment showed that FP-IONP-COOH/DOX had the rule of slowly releasing DOX.4.Investigated of the free radical scavenging ability of the tumor drug delivery system:Nano-water suspensions of each tumor drug delivery system were prepared by organic solvent-mediated method.The measurement results of pyrogallol autoxidation system showed that each nano-water suspension had the ability to scavenge superoxide anion free radicals(O2-·).The methyl violet-Fenton system had verified that each nano-aqueous suspension had the ability to scavenge hydroxyl radicals(·OH)under dark conditions,and also has the ability to generate hydroxyl radicals under light conditions.Further research results showed that FP-DOX had a stronger free radical scavenging ability,which could effectively reduce the toxicity of doxorubicin free radicals. | | Keywords/Search Tags: | Fullerene, Fullerene pyrrolidine, Folate, Ferric tetroxide, Doxorubicin, Nano-water suspension, Free radica | PDF Full Text Request | Related items |
