G. macrophylla Pall. is one kind of the major traditional Chinese medicine, which is first recorded in the "Shen Nong’s herbal classic". The dried root of G. macrophylla Pall.、G.straminea Maxim. 、 G. crassicaulis Duthie ex Burk. and G. dahurica Fisch.,which are all geared to Gentiana L. of Gentianaceae, were regarded as genuine medicine in "Regulations China Pharmacopoeia 2010 edition ". Six mountain region produced Gentiana macrophylla Pall. is a kind of genuine medicinal materials in Ningxia, which used to be wildlife, now planting with large area cultivation after the artificial breeding. However, the research on processing of G. macrophylla Pall. is quite few by now. After returning farmland to forest in the local mountainous areas, the simulation test of Plantation medicine intercropping of Pinus tabulaeformis and G. macrophylla Pall. lack of theoretical support. In this study, taking six mountain region produced Gentiana macrophylla Pall. as the main experimental materials,the change of contents of main components in G. macrophylla Pall. were analysised using the high performance liquid chromatography(HPLC), which was then regarded as the evaluation standard of the quality of G. macrophylla Pall., providing a theoretical basis for the optimization of processing of Gentiana and condition selection of plantation medicine intercropping pattern.The main results are as follows:1.With the root of Gentiana macrophylla Pall. by different origins and different processing from 8 different area segment as materials, Using Chinese medicine chromatographic fingerprint similarity evaluation system(national pharmacopoeia committee),the similarity analysis and fingerprint of root of Gentiana macrophylla Pall. from8 different area segment was established. In order to establish the quality standard of Gentiana and provide theoretical basis for the selection of optimal growth conditions, The optimum conditions of the site for accumulation of effective components of Gentiana macrophylla Pall from Ningxia Liupanshan was analysis by HPLC,taking gentiopicrin and loganin acid as the main factors. The result Indicates that:(1)a total 5 main Common peaks was found in the fingerprints of root of Gentiana macrophylla Pall. from 8 different area segment,which Can be used as the main basis for identification of Gentiana straminea Maxim;(2) According to the clustering analysis, the root of Gentiana macrophylla Pall. from8 different area segmentcan can be individed into 4 categories: Neimenggu Hailaer and Shanxi yangquan, Qinghai Tongren and Neimenggu Tongliao, Hebei Zhangjiakou and Shannxi Taibai, Yunnan Eryuan and Ningxia Liupanshan classified as a category,respectively。2.Selecting Gentiana macrophylla Pall. as material, infiltration water, wetting time,drying temperature, drying time as factors which have three factor levels respectively,and the peak area of Gentiopicroside and Loganic acid in HPLC as index of this experiment.The best combination is the infiltration water 30%,the wetting time 30 min,the drying temperature 30℃,the drying time 30 min.3.Through the investigation and sampling, the different sampling sites in the G.macrophylla Pall.- Pinus tabulaeformis Plantation medicine Intercropping community in Ningxia six mountain region were analyzed. It results that the main companion species are bluegrass and Artemisia argyi; Intercropping of low density results higher content of effective components and plant height; G. macrophylla Pall. prefers cool and humid climate, with higher resistence to cold, covered by bluegrass of large area in the community, belonging to vulnerable populations; The comparison of the effective components of G. macrophylla Pall.from wild, Plantation medicine Intercropping and cultivation is also conducted,which showed that the cultivated G. macrophylla Pall. shared the most effective component content,followed by wild type, the Plantation medicine Intercropping type at least.
