| In consideration of the background of the advance of the human gene project, the researc h of the properties of the DNA molecule is gradually shifting toward developi-ng low cost, ra pid and efficient detection techniques. Recently, The work on DNA sequencing has been achieved successfully with nano-technology, meanwhile, nano-channle technic also show its huge potential on the research on DNA stretching and dynamic analysis, such nano-materials are using on the field of bio-molecules detection more and more frequently, which will be available to achieve the manipulation on bio-molecules on single molecule level, and then to reveal the secret of the whole life body's activity, in order to lay the theoretical foundation on human health.We divide the whole work on three aspects as following:First, stretch DNA molecules on PMMA hydrophobic surface to have a general realize on the character of DNA molecules, and provide an effective stretching method. The work in this part is the preparatory work of the sequencing research on both solution prepare and external condition.Secondly, we fabricated PMMA nano-pillars and micro/nano-channels with the method of electrochemistry and spin coating to duplicate the template. We use such nano-structure surface to study the properties of DNA molecules, and observe the motion state on surface, then research on the mechanism of DNA molecules'action and the forces that have the final decision on the state of DNA.Thirdly, we use AAO templet as the nano-pore structure to detect nano-particles which passing through the pore and analysis the character of such event, all the detection work are under the micro-galvanometer.Finally, we use this AAO templete as the substrate to observe the motion state of DNA droplet on it, and compare it with the state on nano-pillars to study how such nano structures restrain DNA molecules. |
