It is huge challenge for human society in the twenty-first century that the reduce of finite reserved fossil fuel and the continual increase on energy sources requirement and environment pollution resulted in fossil fuel burning. The development of new resources of energy becomes increasingly imperative for realizing the goal of sustainable development. Hydrogen is considered as the best substitute for fossil energy with the advantages of clean combustion, high thermal density and renewability.Photosynthetic bacteria could use sunlight as the source of energy to release hydrogen. Rhodopseudomonas sphaeroides is considered to be one of ideal Photosynthetic bacteria with rapid growth, wide range of utilization of substrate, high utilization of sunlight energy and high environmental adaptability. It is suitable for the research of thesis. In order to offer theoretical foundation and process parameters of application of the hydrogen production of Rhodopseudomonas sphaeroides, application foundation research is the thesis main purpose.The study included following main parts:(1) the optimal conditions for cell growth are micro-aerobic condition, day light, pH6.0-8.0,3000-50001ux and 30-35℃. The main effecting factors in cell growth of Rhodopseudomonas sphaeroides; (2) acetic acid is proved to be cheap and extensive sources through test; (3) the main restrict factors in hydrogen production of Rhodopseudomonas sphaeroides, including self-factors, external factors and contact way between cell and substrate; (4) comparation of hydrogen photoproductivity in free bacterial and immobilized microbes, and analyzed the mass transfer performance of immobilized microbes; (5) anaerobic fermentation combined with Photosynthetic bacteria hydrogen production feasibility study.The experiment results show that:(1) temperature, light intensity and pH is the main controlling factors; (2) the experiment of hydrogen donor source proved the feasibility that acetate is an appropriate hydrogen donor; (3) oxygen and NH4+ restrain hydrogen production; (4) cell age, inoculation amount, nitrogen concentration effect hydrogen production speed; (5) substrate concentration, illumination, temperature and pH effect hydrogen production quantity; (6) the appropriate concentration of Mo ion, Ni ion and Fe ion enhance hydrogen production; (7) use the method of sodium alginate embedding Rhodopseudomonas sphaeroides hydrogen production and specific hydrogen production rate can be increased in various degrees, however, appropriate immobilized carrier need to be seek for, and free cell method is the better way for hydrogen photoproduction; (8) anaerobic fermentation combined with Photosynthetic bacteria hydrogen production is feasible; (9) research approach was given through anaerobic fermentation- Photosynthetic bacteria hydrogen production, that multi-stage reaction can extremely depose organic substance and obtain hydrogen energy.
