B.taibaiensis is the new species of the genus Batrachuperus which was found in Qinling Mountains by Song Mingtao in 2001.The holotype from the upper stream of Heihe River, near Hua Erping Village,Zhouzhi Country, Shaanxi Province, China (33.85°N,107.82°E). In recent years, there are many controversial issues about the distribution of B.taibaiensis in the Qinling Mountains.Qinling Mountains, which traverses the Central China, is the boundary between the north and the south of our country on geography, the watershed of The Yellow River and The Changjiang River water system, intersectant converge and transitional area between the palaearctic region and oriental region. Therefore populations may have divergence on molecular which distribute in the north and south of the Qinling area. Amphibian surveys were made on Qinling Mountains, between the South side of Pingheliang of Ningshan Country, and the North side of Banfangzi and Houzhenzi of Zhouzhi Country, Shaanxi Province, China, from November 2008 to August 2009.We collected to 21 Sample of B.taibaiensis.By analyzing the DNA sequence data and morphological data, gave the following conclusions: (1)MtDNA D-loop region and mtDNA Cytb gene evolved more quickly, and mtDNA D-loop was faster than Cytb gene. Control region are more variable sites, base has already reached saturation, and conversion value is greater than transversions. D-loop was non-coding region, so bases'deletion and insertion was difficult to determine, adopted Cytb gene which was evolved much faster and code, the conclusion was more convincible.(2)By phylogenetic tree defined by 2 genes were divided into 2 clades:the south group of Qinling (Pingheliang), the north group of Qinling (Banfangzi, Houzhenzi).The sample PHL7 could be seen as cryptic species.(3) Morphological data supports the view that B.taibaiensis was divided into two categories by Qinling Mountains, also supports that PHL7 was considered as cryptic species.(4)According to genetic differentiation analysis among 3 population, which suggested that geographical isolation shaped by Qinling Mountains, strapping Qinling Mountains limited gene flowing and diffusion among populations, populations had high genetic differentiation, especially more higher genetic differentiation between the north group of Qinling Mountains and the south group of Qinling Mountains.
