Actinomycetes are a microbe group with high medicinal value which was the first discovered microbe with bio-control effects and have good prospects for development and application in biological control.Through morphology and molecular biology identification,the study revealed that Actinomycete 168 was Microstreptomyces within which no Microstreptomyces strain with plant-virus resisting and antitumor activities have been found presently,and placed a very good foundation for isolating new plant-virus resisting and antitumor substances from Microstreptomyces.This study carried out screening and optimization of fermentation conditions of actinomycete strain 168, passivation effect on TMV was the best when inoculum concentration was 8%,rev was 200r/min,and cultured under 28℃for 6 days.The study also did an antivirus and antitumor test of activity characteristic of active products,results showed that the fermentation products of Microstreptomyces had an inhibition effects on broad spectrum viruses such as TMV,PVY,TuMV and CMV.Passivation rate of crude extract to TMV was up to 84.2%;It was very effective for prevention and treatment to TMV viruses of tobacco and green pepper and had a remarkable proliferation inhibition effect on CMV and PVY,the inhibiton rates reached 74.5%and 69.4%; Through activity test of proliferation suppression of crude extract to tumour cell Hela-60 and 293-T,the inhibitory rate accounted for 37.1%and 28.3%respectively.Study of active fermentation products on Actinomycete strain 168 discovered active substances as two small proteins by using thin-layer chromatography,Sephadex G-75 gel chromatography,ammonium sulfate precipitation on different saturation ranges,ultrafiltration and SDS-PAGE electrophoresis,the molecular weight of the active protein were 34KDa and 22KDa,Concentration of Primary fermentation broth protein wereⅠ168 0.018 ug/mL and 0.25 ug/mL.Activity tests forⅠ168 andⅡ168 demonstrated that the two components both had antivirus and antitumor effects.The experiments provided a series of fundamental dada for searching new resources,researching sustainable application of Actinomycetes and further exploring the potential value of Actinomycetes.
