Sugar beet M14 is one of the monosomic additional line of Beta corolliflora Zoss. in B.vulgaris L., which was obtained from the intercross of the two species (Beta corolliflora Zoss.×B.vulgaris L.)after sequential backcross. M14 constitute of the normal 18 Beta vulgaris L. chromosomes with the No.9 chromosome of Beta corolliflora Zoss.. The result of cytology study showed the chromosome transmission frequency of the monosomic additional line is 96.5%, representing steady and high transmission rate. The embryology study showed high transfer monosomic additional line was a facultative apomict, which reproduced by the diplospory mainly and gamogenesis secondarily. But the DNA molecular genetic evidences were still needed to certificate the apomixis and the homogenicity of the plant lines of M14.Taking Beta vulgaris L. as male parent and M14 as female parent to intercross, using microscope to examine the chromosome number of the filial generation to ascertain whether the filial generation is monosomic additional line, and RAPD was used to filtrate specific male parent strip, the primer was amplified in the parent generation and the filial generation by PCR to examine whether the chromosome recombination occur, and then we used SSR to analyze its heredity relationship, and the heredity characteristic consequently.The specific RAPD sign was transferred to the SCAR sign which is more steady at last. The following main conclusions were obtained.1. The offspring obtained from positive direction intercross were all monosomic additional line, and no result was obtained in the reverse intercross, it may be that the farina of M14 was abortive highly.2. The heredity analysis of the monosomic additional line was used by RAPD and SSR. All the M14 plant accepted the extra farina to reproduce seed generation, and the transmission frequency of the TJ15-3 keeps higher level.3. The SCAR technology was introduced into M14, the technology was established to transfer the RAPD strip to the SCAR, and 3 RAPD signs had been transferred.
