At first,this study aimed to evaluate the effect of polyunsaturated fatty acid on lipid metabolism and inflammatory cytokines in experimental apoE gene knock out mice. The experimental mice were randomized into five groups including control group,group one (n-6/n-3=1),group two (n-6/n-3=5),group three (n-6/n-3=10) and group four (n-6/n-3=68). Every group was feed different diets. After six weeks the mice were executed, and lipids levels of serum and hepatic tissue and serum inflammatory cytokines were detected.At the same time we obtained and observed histological slices.The plasma level of HDL-C,apoAI in experimental group two and three were significantly higher than that of the contol group;A lower apoB level in experimental group and total cholesterol level in the four experimental groups were found compared to the control group. LCAT activity in serum of all the experimental groups was significantly higher than that of the control group, also HMG-CoA reductase show decrease tendency in the experimental group one.The plasma level of IL-2 in all the experimental groups and IFN-γin the experimental group two were significantly lower than that of the control group. Figures of iced slices stained by sudanⅣshowed that the positive regions of aortic wall in the experimental group two and three were smaller than the control group; Iced slices stained by oil red O showed that lipid deposition in all the experimental groups were lower than that of the control group.Different experimental groups impact the atherosclerosis pathogensis from different aspects.Experimental group one is better than other groups according to lipid metabolism result;But from the morphology and inflammatory cytokines results experiment group two is more effective. We also aimed to evaluate the effect of extract by supercritical carbon dioxide extraction from rape pollen on lipid metabolism and inflammatory cytokines in experimental apoE gene knock out mice. The experimental mice were randomized into two groups including control group and extraction group. Every group was feed different diets. After six weeks the mice were executed, and lipids levels of serum and hepatic tissue and serum inflammatory cytokines were detected.At the same time we obtained and observed histological slices. The plasma level of TC,apoAI and apoB were significantly lower in the extraction group than that of the control group;A lower hepatic TC in the extraction group was found compare to the control group. HMG-CoA reductase show great decrease tendency in the extraction group while LCAT activity has no change. The plasma level of IL-2 in the extraction group was lower than that of the control group. Figures of iced slices stained by sudanⅣshowed that the positive region of aortic wall in the extraction group was bigger than the control group; Iced slices stained by oil red O showed that lipid deposition in the extraction groups were lower than the control group.These results suggest that extract from rape pollen enriched in alpha-linolenic acid are effectively in decreasing the levels of lipid in plasma and hepatic tissue to various extent compared to the common diet. This is possibly due to decreasing activity of HMG-CoA reductase and indirectly increasing in DHA concentration of hepatic tissue.The extract also decrease plasma level of IL-2 to regulate atherosclerosis.
