In this work, Protoplasts isolated from cultured cells of S. musstii as a donor were irradiated by UV at the intensity of 380uW/cm~2 for Os, 30s and 1min respectively and were fused with those of Arabidopsis thaliana as a recipient by PEG method.Great amount of regenerated clones were obtained after 3-4 months cultured, some of which differentiated indefinite shoot and root after subcultured on the medium which can induce differentiation. Some of the regenerated clones were identified as symmetric/asymmetric somatic hybrids by using morphological, cytological, isozymic and various DNA-based marker system method such as 5SrDNA space sequence analysis and RAPD analysis. Chromosomes analysis showed that the chromosome numbers of the hybrid clones was 18-22 and there were no chromosome of Arabidopsis in both symmetric and asymmetric somatic hybrids clones. RAPD analysis showed that all of the hybrid clones had 83%-87% nuclear DNAs of S. musstii and the nuclear DNAs of Arabiaopsis thaliana only account for 12%, which suggested that the nuclear genome of the hybrids biased to the donor S. musstii. SSR analysis revealed that the hybrid clones had bands and some had new bands. GCMS analysis of methanol-soluble components metabolism grams demonstrated that most hybrids clones had the chemical substances of some novel compounds, which manifested that the metabolism related genes of S. musstii were transformed to the hybrids and expressed. The result further confirmed that somatic hybridization was an effective way to transfer useful genes.Protoplasts of Triticum aestivum L. as a recipient and R.ongneria amurensis as the donor were fused by using PEG method.
