A huge challenge to plant research is to understand how the plants alert himself facing stress of environment and the relation among the networks that underlie plant stress and defense responses. The results of most studies suggest that plant stress tolerance is usually correlated with a more efficient antioxidant system. Therefore, study the oxidation stress response in plant with the aim of enhance the plant stress tolerance has agricultural and economic importance.In the post-genomic era, technology development for large scale protein analysis looms large Reductionism methods, such as studying one gene, one protein or one pathway in an organism, have contributed greatly to our understanding of many basic principles of life. However, a comprehensive picture of biology will be difficult to grasp until more integrative approaches are utilized. Recently, the significant accomplishments of genomics and proteomics are making the large scale protein analysis of all expressed cellular components into reality [2-4]. Using proteomics systemic researching the antioxidation mechanism of the plant can help us to understand the response network of plant in stress.In our study we applied a proteomic strategy by using two dimensional electrophoresis and MS to investigate the oxidation stress responsive proteins in Arabidopsis roots. The thesis tries to explore relevant parameter to establish a 2DE and SCX-LC-MS/MS technique platform for the study in proteomics of Arabidopsis root.
