Qin Ba mountain region was very closed in the past, thus forming a relative separated population. There are many children with mental retardation (MR) in the region and familial aggregation is common. This suggests that genetic factors may also be very important in the forming of MR, except environmental factors. Our research work have investigate the possibility of the association of IL1RAPL1 gene with children's intelligence.In Qin-Ba mountain area, studies were carried out among children from two different areas Zha Shui and An Kang two experiment centers. The blood samples were form 596 children ,inclouding children were of low intelligence, children were of marginal intelligence, and the other children were of normal intelligence. DXS1218 and DXS9896 that lies in ILIRAPLI gene were used as genetic marker to investigate the allele distributions and make association analysis with intelligence character.Blood samples were treated with anticoagulant by using EDTA, and genomic DNA was obtained by phenol-chloroform extraction. Various methods, such as PCR, PAGE and DNA sequencing were carried out in the following analysis. The results showed that DXS1218 has 17 alleles, and there was no difference in the distribution among An Kang and Zha Shui(x~2=22.980, p>0. 05),but the difference between male and female is very significant (x~2=45. 091, p<0. 01). The allelic type frequence of DXS1218 in all children was in accordance with Hardy-Weinberg equilibrium (P>0.05) . The heterozygosity of DXS1218 is 0.8988. DXS9896 has 11 alleles, and there was no difference in the distribution allele frequence between An Kang and Zha Shui (x~2=8. 929, p>0.05),and also among different gender(x~2=11. 499, p>0. 05).The allelic type frequence of was in accordance with Hardy-Weinberg equilibrium (p>0.05). The heterozygosity of DXS9896 is 0.8678. Thus, the DXS1218 and DXS9896 can be used in relative research of genetics.The Chi-square test of the association among groups with different intelligence,was carried out. All the results indicated that, the DXS1218 allele A13-A15 in male individual and the DXS9896 allele gene B2, B7 in all sample is associated with normal intelligence, and DXS9896 allele gene B4, B5 is associated with low intelligence. The results suggested that there were many different loci in IL1RAPL1 which were exerting different effects on intelligence development. Ourfindings provide evidence and inspiration for the further study in the future.
