RNA interference (RNAi) is a rapidly emerging and powerful technique used to investigate gene function by degrading a specific mRNA target in a cell or organism and thus silencing or knocking down the level of the encoded protein. According the conservative domain of Arabidopsis thaliana FIE cDNA which was published in GenBank, designs 4 pairs of 29nt oligonacleotide, transcribes and synthesises 4 kinds of siRNA which will be used in interference. After interference, collects the sample of 1, 2, 3, 4day. Using the FIE probe which was coloned from Beta vulgaris M14, hybridizes the total RNA of flower bud. According the strongor weak signal, identifies the function of RNA interference.And according the sequence of FIE gene,designs a pair of primer, uses RT-PCR and colones the specific fragment of Beta vulgaris M14 FIE gene.According the RT-PCR production identifies the function of RNA interference. According the result, after synthesising siRNA get into the plant, interference is finded and after 3 days thefunction of interference is obvious which will last little time.
