Intracytoplasmic sperm injection(ICSI) is a very important assistant reproductive technology for treatment of man infertility and fundamental research , Success in ICSI in human and other animals has recently been achieved., The present pilot studies were conducted to establish a preliminary procedure for ICSI in mice, explore the experimental conditions of ICSI in mice, investigate factors influencing the test results, and accumulate technologic basis for the next studies on pronuclear exchange in mice .The mature oocytes were collected from superovulated female mice and were treated in washing solutions Mice fresh sperm was injected into an oocyte byassistance of a micromanipulation system?The development of injected oocytes was examined at different stages constantly. Because mice oocytes are easy to be damaged by the mechanic stimulation, the survival rate of injected oocytes is very Iow0 In this experiment, the survival rate is about 25%, the two-cell rate is 50%, and the blastocyst rate is about 24%.During the course of mice ICSI,some critical factors were realized on influencing the experimental results, including micromanipulation systems, diameters of injection pipettes, concentrations of PVP and positions of the first polar body located in oocyte atlCSL...
