Implantation,which is initiated when the recognition and direct interaction occur between a hatched free embryo with the surface of uterus, is a key step in reproduction. Successful implantation results from a coordinated series of event that would allow establishment of a timely dialogue between a receptive endometrium and an intrusive blastocyst. Many factors involved in the process may consist of the regulation net of implantation. The interactions among those of implantation-related factors regulate embryo growth and implantation process meticulously and coordinately. ECM and some cell factors play important roles during the process.Oligosaccharide LeY is stage specifically expressed on the cell surface of the endometrium and blastocyst during implantation stage of mice. The results obtained from our previous work in vivo and in vitro implantation models showed that implantation is significantly inhibited by masking LeY on the surface of embryo and/or epithelial cells with its specific AH6, and meanwhile the secretion and the expression of MMPs and EGF were also significantly declined. All the date support that LeY may function not only as a mediator at the initial stage of implantation, but also maybe as a signal molecule involving the regulating net to effect other implantation relating factors.To identify LeY antibody may involved in the implantational regulatory net. In this dissertation, the effects of LeY were studied by blocking LeY of embryo with specific monoclonal antibody AH6 in vitro; By using of RT-PCR and immuno-dot blots, to detect the secretion and gene expression of implantation-related factors, including FN, LN, MMPs,TGFβ1 and VEGF , and further explore the interactions among LeY, MMPs,and the major component of ECM, which play important roles in embryo imbedding , involved implantation regulationnet.The results in this dissertation was summarized as follows:1.oligosaccharide LeY blocked by AH6 antibody, the secretion of FN and LN in blastocysts are gradually increased, which suggests that LeY may play a role in regulating the secretions of FN and LN of murine blastocyst at pre-implantaion stage. At the same time MMPs secretion and gene expression of blastocyst are significantly inhibited. It idicates that there may be connected between the alteration of MMPs and ECM induced. It may caused by the reducing of MMPs activity and therefore the content of LN and FN are induced by their degradation decreased 2. The expression and secretions of TGFβ1 and VEGF in embryos are significantly inhibited by LeY antibody, which suggested LeY may be involved regulation of implantation,by affecting the expression and secretion of TGFβ1 and VEGF.
