Using heat treatment method and Ion-exchange chromatography, we separated and purified porcine kidney aminoacylase and got ideal result. We dedicated to get the relationship between pH value and the retention rate of aminoacylase activity during lyophilization.We studied the influence of some factor such as temperature.time.Co2~ concentration and operation pattern on raw separation of aminoacylase from porcine kidney with heat treatment.We knew that the best operation mode is to heat porcine kidney aminoacylase raw extraction solution for about 30 mm under 70C with unceasing blendering in which the Co2~ concentration is 1 Onimol per liter. Comparing with traditional cold acetone deposition, we found that heat treatment method is better than traditional method in all ways (e.g. Recycling rate, purification rate and operation pattern) all which suggest the industrial prospect of heat treatment method.We emphasized on tracing the best pH value of elution solution in porcine aminoacylase purification with Resource-Q 15 Ion-exchange chromatography. We conih-med the best pH value of elution solution to be 7.8.Purity of aminiacylase got through this method was proved by different means.We investigated the arninoacylase activity changing at different pH value along time and found that the protein molecule may be under "transition state" in some special condition. Under "transition state ", we studied the relationship between aminoacylase activity retention and pH value and got some result disagreeing with those result from reference. We suggested an acceptable explain for that result.This new method used in this paper for porcine kidney aminoacylase purification has both theoretical and practical significance. Our research work on the relationship between pH value and enzyme activity retention during lyophilization offered a new view of protein denaturation during lyophilization.
