Abiotic stresses such as drought, salinity and low temperature is the most important factor to restrict crop yield and quality. It is a great significance to cultivate new varieties of crops in strong resistance to enhance agricultural productivity and save resources. Transgenic Breeding is an effective way to improve the resistance of crops. A number of transcription factors play an important role in the regulation in plant response to stress. Because a transcription factor can regulate the expression of a number of stress-related genes, it may comprehensively enhance the resistance of crops through genetic transformation to improve resistance of breeding crops.AP2/EREBP which is specially possessed by plants is a class of transcription factors. Many members of the family play an important part in the process of the resistant reaction of plants. HRD transcription factor which is a regulating protein produced by plants in the process of the response to drought stress is found recently. It is a member of the transcription factor for AP2/ERF-like subfamily and can improve WUE of plants in the condition of drought.The HRD gene does not have introns, so we cloned the gene HRD from the Arabidopsis genome by the method of PCR. Then pBIN-HRD was constructed using the plant expression vector pBIN438 as the backbone through enzyme digestion and ligation to transform Agrobacterium tumefaciens GV3101.Tomato were infested by Agrobacterium tumefaciens GV3101containing the plant expression vector pBIN-HRD. Regenerated tomato plants were obtained after coculture, successive induction of the indefinite buds and roots produced. PCR and RT-PCR analysis indicated that the foreign gene HRD had been integrated into the genome of tomato and acquired expression. The analysis of chlorophyll content, water conservation experiment of leaves and drought resistance induced by PEG were done to transgenic tomato. The results demonstrated that HRD gene can improve the drought resistance of tomato to some extent. At the same time, tobacco were also infested by Agrobacterium tumefaciens GV3101containing the plant expression vector pBIN-HRD in this study and obtained expression.In this study, we cloned HRD gene from the Arabidopsis, transformed it into tobacco and tomato using the Agrobacterium tumefaciens mediated method and analysed the resistance of transgenic plants, which is very significant to resistance breeding of plants.
