| Paecilomyces hepialid is one of the anamorph of Cordyceps sinensis and itseffective ingredients is simiiar to natural Cordyceps sinensis. The study on it is ofgreat significance. We use chemometric methods and satisfaction function to improvethe fermentation level of Paecilomyces hepiali SY02which was bred by thelaboratory. First of all, the type and concentration of Paecilomyces SY02fermentationmedium carbon and nitrogen sources and inorganic salt were selected by single factormethod, then key medium components were identified by Plackett-Burman design(PBD) and further optimized by Box-Behnken design (BBD). The best culturemedium formula of obtained through the neural network and genetic algorithmanalysis is: glucose3.35%, peptone1.686%, yeast extract1.80%, KH2PO4·3H2O0.20%, MgSO4·7H2O0.40%, ZnCl20.002%and VB10.01%. A mean value ofdesirability values Dv=0.5124was obtained, which was41.925%higher than thevalue achieved by the basal medium.Plackett-Burman design was used to identify the key components of5Lfermentation conditions on the yield of polysaccharide. The stirring speed was thesignificant factor with the optimum value of300r/min. The suitable culture conditionwas: initial pH natural, temperature26℃, culture time4d, seed age3d, rotatingspeed300r/min, inoculation amount7%, liquid volume in5L flask3L. The biomass,the production of adenosine, the polysaccharideã€the cordyceps acid and protein yieldswere15.09g/Lã€0.07g/Lã€1.04g/Lã€1.81g/L and9.40g/L, respectively.The quality standard of Paecilomyces hepiali powder was studied on the basis ofabove,the character identification, thin-layer chromatography, gene identification method was used for the qualitative identification of samples, the determination ofmoistureã€ash contentã€arsenic salt and heavy metals was carried on. The moisturecontent of not more than7%, total ash content of not more than1.3%, acid insolubleash content of not more than0.15%were planed to put in the check items of thesample quality standard. HPLC method was used to determine the adenosine content,tentatively the productadenosine content shall not be less than2.53mg/g.Preliminary study on immune activity of SY02fungus powder confirmed that thepowder can enhance the non-specific immune function of mice by raise the level ofNK cells killing. Secondly, the fungus powder can make the delayed typehypersensitivity reaction aggravated, lymphocyte transformation rate increasedproving that the powder can improve the cellular immune function of organism.Paecilomyces hepiali powder has raising effect on IL10, IL12, IL12R, and IFN-α,further experimental studies need to be done to explain the exact mechanism of theaction. |
