Panax notoginseng is a valuable herbal medicine of our country tradition. The microbial fermentation of Panax notoginseng can produce higher physiological activity and more valuable application products, and it has good application prospects.This study used three kinds of Cordyceps fungui (Paecilomyces hepiali Chen&Dai, Paecilomyces militaris Liang and Paecilomyces cicadae Samson) to ferment the liquid culture medium with raw Panax notoginseng, detected the content of active components in fermentation product. HPLC was used to make a qualitative and quantitative analysis for the changes of saponins monomer composition among the process of fermentation. HPLC was used to make a quantitative analysis for cordycepin and adenosine. The colorimetric method was used to measure mannitol content of fermentation product. The results showed that ginsenosides Rd in the fermentation product by Paecilomyces hepiali and Paecilomyces militaris were4times as much as before, thus achieved high yield of rare ginsenoside Rd. At the same time, they contained adenosine, cordycepin and mannito.Paecilomyces hepiali, Paecilomyces militaris and Paecilomyces cicadae were used to ferment raw Panax notoginseng in solid media, and the active components in fermentation products were detected. The three kinds of Cordyceps fungi could grow well in the culture media. Ginsenosides Rd in fermentation product of Paecilomyces hepiali and Paecilomyces militaris achieved high yields which were5times as much as original medicinal materials of sterilization. The contents of adenosine and mannitol in the fermented products corresponded with those in wild Cordyceps sinensis. Cordycepin content of fermented product in Paecilomyces militaris was868.53μg/g, which was far higher than that of wild Cordyceps sinensis. When Panax notoginseng was used as substrate, the technologies of solid fermentation were optimized using total saponins, ginsenoside Rd, adenosine, cordycepin and mannitol as markers. The best fermentation times of Paecilomyces hepiali, Paecilomyces militaris were16d and20d, respectively. Meanwhile, in solid fermentation product of Paecilomyces hepiali, ginsenoside Rd were1.25times as much as those of the liquid fermentation product, total saponins and adenosine corresponded with the liquid fermented product, but mannitol was lower than that in liquid fermented product. In solid fermentation product of Paecilomyces militaris, ginsenoside Rd and cordycepin were were1.33times and18.87times as much as those of the liquid fermentation product, but total saponins, adenosine and mannitol were lower than those in liquid fermented product.With silica gel column chromatography and recrystallization method, total saponins were extracted from Panax notoginseng. The culture medium and time for Paecilomyces hepiali and Paecilomyces militaris were optimized. TLC was used to analyse the transformation of ginsenosides Rb1in fermented products. The results showed that Paecilomyces hepiali and Paecilomyces militaris had the ability to convert Rbl to ginsenosides Rd, and the intracellular enzyme catalyzed the reaction.This study fermented Panax notoginseng using Cordyceps fungi, and the fenmented products contained rare saponins, adenosine, cordycepin and mannitol.It was possible that the medicinal ingredients of Cordyceps fungi and Panax notoginseng could be synthetized at the same time in once fermentation process. Based on the results, rich and new medicinal resources can be got with the interaction between medicinal microorganisms and plants drug.
