Green walnut husks is an immature ericarp of Juglans mandshurica Maxim,Which belong to the Juglandaceae family, Juglans genus. It is widely distributed in east and north region of China. In addition to walnut nuts, bark fruit, leaves the root bark also can be used as medicine, various parts of the extract as folk medicine has many years of history, walnuts plants have antioxidant, anti-tumor and significant insecticidal activity. In this paper, the chemical composition and insecticidal activity of green walnut husks is studied, mainly includes the following tasks:1> This experiment separated and isolated alcohol extract of green walnut husks by means of silica gel, ODS, MCI, Sephadex LH-20, HW-40C column chromatography, combined with physicochemical properties and modern spectroscopy,15compounds were identified:1(2H)-Naphthalenone,3,4-dihydro-8-hydroxy-4-butoxy (1), Myricananin F (2), stigmast-5-en-3β,7α-diol (3), stigmast-5-en-3β,7β-diol (4),3β,23-dihydroxy-Olean-12-en-28-oic acid (5),3β,23-dihydroxy-Urs-12-en-28-oic acid (6), n-Butyl phthalate (7), corosolic acid (8), arjunolic acid (9), olenolic acid (10),β -sitosterol (11), daucosterol (12), regiolone (13),4,5-dihydroxy-α-tetralone (14),3,4-dihydroxybenzoic acid (15). Compound1is a new compound, compound2,3,4,5,6were isolated from Julans genus for the fist time.2〠Phenol oxidase as a target, investigated enzyme inhibition by Juglone,1,4-naphthoquinone, menadione and chloroform extracts, juglone inhibite phenol oxidase activity significantly, in0.1mol/L phosphate buffer solution (pH=6.80), at37℃, led to a50%decrease in enzyme activity required Juglone concentration (IC50) of0.832mmol/L.3-. Applications of NMR-Based Metabonomics on the studies of Juglone’s insecticidal activity:With the help of Partial Least Squares (PLS) model analysis, analyzed metabolites components of aphid hemolymph from the control group and treatment group, such as:glucose, trehalose, betaine, valine, lactate, alanine, dimethylamine, taurine, putrescine, etc. The changes in metabolitesl can be used as potential Markers of insecticidal activity of Juglone and provide valuable clues to our in-depth understanding of the pesticides’toxicity mechanism. |
