Monascus was a kind of fungus, could produce the secondary metabolites of monascus pigment in its growth process. As the natural pigment it was used to replaced by synthetic pigment increasingly.Monascus pigment not only could be used as natural pigment, but also had inhibition of lipase activity. Currently, some weight-reducing aid affected brain to suppress appetite and others by inhibiting the activity of lipase to reduce weight, but all of them were synthetic, it had different degree side effects. So more and more scholars would research natural drugs.This paper mainly through adding D-penicillamine in process of Monascus fermentation to induce lots of monascus pigment, improving the efficiency of monascus pigment to inhibit the lipase activity, and optimize the conditions of induction. At last, to separate and extracte the monascus pigment, getting the single pigment of deepest colors and the best inhibition to lipase activity.By the single factor method to optimized the culture medium composition and culture conditions of Monascus, to get more mycelium and pigment. The optimal component of culture medium was as follows:glucose2.5g, sodium nitrate1g, potassium dihydrogen phosphate0.075g, magnesium sulfate0.05g and distilled water50mL. The optimal of culture conditions was as follows:the initial pH of culture medium was3.8, the temperature was30℃, the incubation time was12days, the bottling quantity was50mL/250mL.Optimzed the add time and adding amount of inducer. The result was as follows:after sterilization of culture medium added the inducer was best, adding inducer before the sterilization had not effect, and after5days the effect was unconspicuous. The adding quantity of inducer was0.042%o. In this condition to culture Monascus produced pigments, the colour value was best, and it could obvious inhibit the lipase activity.In the best condition to culture monascus pigments, to extract and segregat it, the obtained material for TLC detection, obtain7single pigments, dry its and give numbers to1-7. Determined them color value and inhibit efficiency to lipase activity. The result was the6number monascus pigment’s color value was best and had distinct inhibit efficiency to lipase activity. Others monascus pigment’s color value was lighter and basically had no effect on lipase activity.
