In this paper, Zizyphus jujuba cv. Junzao of Alar as experimental materials we studied the extractionprocess of water-soluble polysaccharide, separation and purification technology, purification and purityidentification classification, monosaccharide composition and in vitro antioxidant activity.1With response surface determine the optimum parameter of hot water extraction of Junzaowater-soluble polysaccharide process conditions.The results showde that: the order of the factors affectingtemperature> Time> solid-liquid ratio> times, the best parameters for the extraction time90min, extractiontemperature89℃, liquid-solid ratio29:1, extraction times2. Under optimum conditions, the extraction rateof polysaccharides Junzao up46.42percent.2We removed Junzao polysaccharide protein with the method Sevage, trichloroacetic acid andhydrochloric acid, and use orthogonal experiment to optimize the best process conditions of removal ofprotein.The results showed that: Sevage law deproteinized better; After optimization of process conditionsfor the oscillation time of25min, chloroform: n-butanol3:1sample: Sevage reagent (chloroform-butanol)5:2, adding the number of to six times. Under optimum conditions, the polysaccharide retention rate90.44%±1.15%, protein removal rate was87.76%±0.82%.3Macroporous resin to remove pigment Junzao polysaccharide and with orthogonal experiment tooptimize the best bleaching method of process conditions macroporous. The results showed that: thesample flow rate of1.0mL/min, the sample volume was20mL, the sample solution pH of6. Underoptimum adsorption conditions, the verification test, get polysaccharide adsorption was85.62%±1.27%,pigment absorption rate of86.59%±2.41%. Through dynamic analysis experiments, elution rate reached1.0mL/min, the desorption rate showed a maximum polysaccharide desorption rate83.54%±0.80%,pigment removal rate was83.02%±0.47%. When eluent consumption reached60mL, desorption rate wasmaximum, polysaccharides desorption rate83.81%±1.06%, pigment removal rate was83.26%±1.63%.4By the DE-32cellulose column chromatography on Sephadex G-100column grading, The resultshowed that: JWP-1polysaccharide content was96.81%±1.25%, as a pale white fibrous solid; JWP-2polysaccharide content was98.96%±1.52%, to light brown powder; JWP-3polysaccharide content was97.62%±1.42%, as a white powder; JWP-4polysaccharide content was95.54%±1.39%, as a light tanpowder; JWP-5polysaccharide content was97.49%±1.12%, as a white powder. And by UV spectroscopy,and each component in the260nm absorption peak at280nm were not, indicating that no protein andnucleic acids.5The result of infrared spectroscopy of Junzao polysaccharide JWP-1, JWP-2, JWP-3, JWP-4, JWP-5showed that in the vicinity of3700-3200cm-1,2970-2840cm-1,1420,1200-1000cm-1characteristicabsorption peak, indicating a hydroxyl group, a carboxyl group having a polysaccharide structure. In the936cm-1of these properties are no peaks, indicating that contain furan ring. In the absorption peak at870cm-1for the deformation vibration of β-D-pyranose of CH. Relative molecular weight of approximately:66.2kDa,81.2kDa,76.3kDa,86.4kDa and139.6kDa. Contain mannose, rhamnose, glucose, galactose,arabinose, indicating Junzao polysaccharide is composed of different monosaccharide composition,structure and diverse heteropolysaccharide.6With spectrophotometry determined Junzao polysaccharides on DPPH radical, hydroxyl radical, ABTS+radicals and superoxide anion radical scavenging, and reducing power and chelating ability. Theresults showed that: Junzao certain polysaccharides on free radical scavenging capacity; and within theconcentration range tested, a dose-response relationship; the higher the free radical scavenging ability ofthe polysaccharide content of the stronger; polysaccharides possess the ability of deoxidation and chelatingcapacity, and can be use as the iron chelators.7Polysaccharides possess restraint of activty of Tyrosinase, and the restraint is different withpolysaccharides from different component, and the best is JWP-5(IC50=1.43mg/mL),which can be usedas in medicine inhibitor。...
