| Lignin raw material, is the most abundant renewable biomass in the world. It can be converted to fermentable sugar after the pretreatment, enzymatic hydrolysis. Using biomass-based raw materials for biconversion, It holds a great hope and future to solve energy crisis for its resource saving and environment protecting.23-Butanediol is an important kind of widely used chemical material and new fuel, but has not been able to achieve mass production. the production of2,3-butanediol by biotransformation, has been the subject of for scholars.Based the "liquefaction"technology in the lab. The product of liquefaction of straw (mainly cellulose and hemicellulose) were digested by the enzyme through a series of experiments to determine the best solution.we find the liquefaction not only improves the Cellulose content,but also breakes the crystal structure between the Lignin raw material, reduces the amount of enzyme and hydrolysis time, improves the rate of hydrolysis rate and the amount of reducing sugar. We selecte strains Klebsiella, optimized culture conditions,to determine2,3-butanediol the best fermentation conditions. Using of enzyme solution as C source, we achieve good results about the2,3-butanediol fermentation finally.We conclude:the best hydrolysis conditions were:solid to liquid15:100, enzyme concentration68.8IU/g, pH4.8,50℃, rotation speed200r/min, hydrolysis time is108h,the final glucose concentration is75.5mg/mL, xylose10.2mg/mL, the yield of reducing sugar hydrolysis is56.7%; fermentation conditions:glucose100mg/mL, temperature37℃,10%inoculum, medium volume was80ml, rotation speed160r/min, pH5.6, then2,3-butanediol the highest concentration To39.8mg/mL, about80%theoretical conversion rate; use the fermented hydrolyzate glucose concentration100mg/mL.we obtaine2,3-butanediol concentration27.4mg/mL. |
