In the present research both flocculation method based on the water extraction and two-aqueous phase extraction are adopted to perform the extraction and purification of chlorogenic acid in honeysuckle. In order to solve this problem we try to employ the modified chitosan, to defecate the extract, and by optimizing the process conditions through the experiment flocculating effect becomes even more significant. The flocculating ration can be up to5.334%and transmittance89.5%under the condition that the amount of flocculant is0.5g/50mL, the temperature is46℃, the flocculation is20min, pH value is5.0, and stirring rate is500r/min. The experiment indicates that better clarifying effect is achieved and at the same time less quantity of chlorogenic acid is lost compared with the traditional water extraction method.With the advantages of high biocompatibility, mild condition, easy amplification, and so on, the two-aqueous phase extraction is a new type of biochemical separation technology. Through investigating the pase ability of inorganic salts and organics, those inorganic salts and organics with strong phase ability are chosen as the constituents of the system. By inspecting the split phase ability we get the result that ethanol, acetone, and isopropyl alcohol can not constitute the two-aqueous phase extraction with potassium dihydrogen phosphate. Among the above three organic phases ammonium always has stronger split phase ability than potassium dihydrogen phosphate. In different inorganic salt solutions, the split phase abilities of the three organic solvent can be shown as ethanol>acetone>isopropyl alcohol. Therefore, ethanol/ammonium sulfate should be taken as the constituents of the two-aqueous phase extraction.The experiment of ethanol/ammonium sulfate two-aqueous phase extracting chlorogenic acid is conducted under the condition that mass fraction of ethanol is30%, mass fraction of ammonium sulfate is20%, and pH value is5.0at room temperature. With the advantages of distribution coefficient61.11, extraction ratio of chlorogenic acid76.82%, purity of the product71.46%and being free from contamination, the two-aqueous phase extraction is far better than the traditional liquid-liquid extraction method. This method has not been reported yet.
