1,3-diacylglycerol,as a muhifunetional nutrimental additives,has a extensive application prospect in food industry,pharmaceutical industry,chemical engineering industry and so on. Its preparation method can be divided into chemical method and enzymatic method. Chemical method is multifarious, when the conditions of enzymatic method is mild,is friendly to environment, and the quality of products is very good. Fermentation tank can achieve continuous preparation. The preparation process of1,3-diacylglycerol were introduced,and its industrial preparation was simulate studied.1ã€The triacylglycerol of cow milk fat was hydrolyzed by the catalysis of lipase to fatty acid.Hydrolysis parameter,such as reaction time,temperature,enzyme dosage and water amount were investigated.Based on single factor experiments, response surface methodology was used to explore the effects of hydrolysis temperature, enzyme dosage and water amount as well as their cross-interactions on the hydrolysis of triacylglycerol. Through the regression analysis, the optimal conditions of hydrolysis were obtained as follows:reaction temperature50.7℃,enzyme dosage0.8%,water amount26%.Under these conditions,the acid value (NaOH) of the hydrolysate was35.98mg/g.2ã€Through the effective integration of enzymatic hydrolysis reaction and enzyme catalysis esterification reaction, the enzymatic preparation of1,3-diacylglycerol has been proposed. The esterification reaction between fatty acids and glycerol was mainly studied. The fatty acids obtained from the hydrolysis of triglyceride was used as reaction substrate. The esterification reaction between fatty acids and glycerin was proceeded, under the catalysis of Lipozyme RM IM,1,3-diacylglycerol was generated.The influences of factors on reaction, including the reaction temperature,amount of enzyme,molar ratio of fatty acid to glycerol,time,were decided. Based on single factor experiments, response surface methodology was used to optimize the effects of each factor as well as their cross-interactions on esterification reaction. Through regression analysis, get the best esterification process parameters:the reaction temperature:55℃, amount of enzyme:3%,molar ratio of fatty acid to glycerol:1.5:l,time:12.5h,the content of1,3-diacylglycerol:45.13%.3ã€Triglyceride and glycerol were used as reaction substrate, glycerol solution reaction was reacted, under the catalysis of Lipozyme RM IM,1,3- diacylglycerol was produced.Fermenta.tion tank was used as biological reactors, the influence of different amount of substrate on content of1,3-diacylglycerol was studied,but time, temperature, amount of enzyme, amount of water was constant The result show that:when the other factors keep constant, the content of1,3-diacylglycerol in reaction product is steady though add the amount of reaction substrate. The optimum condition as follows:the reaction temperature:51℃,amount of enzyme:6.4%,molar ratio of the substrate:1.3:1, time:16.5h,the content of1,3-diacylglycerol in reaction products is between33%-34%. Scale of production was successfully simulated in the laboratory. Provide technical parameters and solutions for industrial production. |
