Cysteine and Thiol compounds which contains Cysteine (referred to as Thiol compounds) play significant biological functions in the creatures, and most of them distribute in the plant cells. Thiol compounds like GSH, Cys and others are important healthy factors in the organism. The content distribution of Thiol compounds can be decreased by organism senility and certain diseases. One of the most important way to prevent diseases, cure disease and prolong life is is to increase the content distribution of Thiol compounds through selecting the plants of medicine and food as the exogenous supplements and being absorbed by human body directly. But now the plant samples are full of problems, such as missing detection, improper methods, lack of information of Thiol compounds content, and selections of different Thiol compounds contents and so on. These bring out many problems to the selecting of the plants of medicine and food and the evaluation of their biological functions contents. This paper inspects the Thiol compounds in lotus seeds and hovenia dulcis thunb detemined by "in-situ" derivatizations using high-performance liquid chromatography and spectrometry.Dertermination of GSH and total Thiols in lotus seeds using HPLCHPLC method can be used to measure various thiol compounds in biological samples. In this research, the content of GSH and total Thiols in lotus seeds determined by "in-situ" derivatizations choosing DTNB as the derivatization reagent. During the process the detecting the total Thiols, it is not possible to get the exact qualitative analysis of the total content of Thiol compounds because of the tailing phenomenon of chromatograpic peak of its derivative products. Then optimize the experiment by selecting the tetrahydrofuran and methanoic acid. Finding that according to certain ratio (9:1:0.1), adding to the tetrahydrofuran and methanoic acid can eliminate the tailing in the chromatograpic peak and establish HPLC to get the GSH and total Thiols. The content of GSH and total Thiols in lotus seeds are0.9711μmol/g and1.3686μmol/g, the recovery rate are101.52%and99.93%, the detection limit are4.21μmol/L and4.63μmol/L.Dertermination of GSH and total Thiols in lotus seeds using spectrometryGSH and total Thiols in lotus seeds are dertermined by using UV-Vis Spectrophotometry and fluorescence Spectro-metry with "in-situ" derivatizations. The content of GSH and total Thiols in lotus seeds are0.9443-0.9576μmol/g and1.3267-1.3452μmol/g, the recovery rate are101.35%and100.32%, the detection limit are1.365μmol/L and0.401μmol/L.Dertermination of GSH and total thiols in hovneia dulcis thumb using spectrometry4-DPS derivatization reagent is similar to DTNB, can be used to measure total thiols like small molecular thiolsl and protein thiols in the samples. GSH and total thiols in hovneia dulcis thumb are determined by using UV-Vis spectrophotometry and fluorescence spectrometry with4-DPS and OPA derivatization reagent. The content of GSH and total thiols in hovneia dulcis thumb are0.08401-0.08582μmol/g and2.391-2.462μmol/g, the recovery rate are101.72%and99.25%, the detection limit are0.02377μmol/L and0.03641μmol/L.
